奥曲肽对人胃癌细胞抑制作用的实验研究

目的 探讨生长抑素类似物奥曲肽(OCT)对胃癌细胞株MKN_(45)生长的体外调节作用。方法 MTT法测定OCT对MKN_(45)细胞增殖的调控;流式细胞术分析OCT对MKN_(45)细胞周期分布的影响;RIA法测定表皮生长因子(EGF)、转化生长因子-α(TGF-α)含量的变化。结果 OCT对MKN_(45)细胞生长有抑制作用,以在1×10~(-6)mol/L浓度时最明显;OCT可诱导MKN_(45)细胞G_0/G_1阻滞,加入OCT后24 h最显著,同期G_2/M期细胞比例亦减少;OCT能抑制胃癌细胞合成EGF、TGF-α的能力。结论 OCT对胃癌生长具有负性调控作用,为胃癌患者应用OCT治疗提供了依据。

EXPERIMENTAL STUDY OF INHIBITION OF OCTREOTIDE ON HUMAN GASTRIC CANCER CELLS GROWTH IN VITRO

Objective: To study the regulatory effects of somatostatin analogue(octreotide, OCT) on the growth of gastric cancer cell line, MKN_(45) in vitro. Methods: Cell proliferation was evaluated by means of MTT assay, and cell cycle distribution was measured by flowing cytometric analysis. The contents of EGF and TGF-αwere determined by radioimmunoassay(RIA). Results: The growth of human gastric cancer cell lines MKN_(45) were inhibited by OCT. inducing MKN_(45) to G_0/G_1 arrest, which peaked at 24 h. At 24 h and the percentage of G_2/M cells also decreased as compared with the controls. The contents of EGF and TGF-α were significantly inhibited by OCT. Conclusions: OCT inhibits the growth of gastric cancer cells which provide an experimental foundation for the patients of gastric cancer to use OCT.
