抑颤汤对帕金森病模型大鼠行为学及脑黑质细胞形态学的作用

摘    要:背景目前治疗帕金森病(Parkinson disease,PD)仍以多巴胺替代疗法为主,但因其副作用多,有效时间短等弊病,使神经保护性治疗的探索日益受到重视,而中药治疗具有良好的效果且不良反应较少,探讨其作用机制将有利于临床应用.目的观察抑颤汤治疗 PD在保护和促进黑质细胞修复方面的效果及其相关机制.设计完全随机对照实验研究. 地点和对象解放军总医院医学实验动物中心,实验动物为SD大鼠,分为正常组、生理盐水组、抑颤汤组. 干预 6-羟基多巴胺诱发建立PD大鼠模型,抑颤汤组大鼠灌胃法给药,生理盐水组和正常组生理盐水灌胃.给药后第5周开始观察大鼠行为学改变,连续观察 4周,给药结束后,各组大鼠断头处死,制备大脑切片,经苏木精-伊红染色后,用光学显微镜及电子显微镜观察脑黑质细胞形态的改变.主要观察指标各组大鼠行为学改变和脑黑质细胞形态. 结果造模后经阿朴吗啡诱发,正常组大鼠旋转次数平均为22圈 /40 min,无固定旋转方向;造模组大鼠旋转次数平均为 343圈 /40 min,两组相比差异有显著性意义(t=3.390, P< 0.001);治疗后,抑颤汤组大鼠旋转次数为 340圈 /40 min,两组相比差异有显著性意义(t=2.648, P< 0.01).病理形态学观察,抑颤汤组脑黑质细胞数量较生理盐水组明显增多,神经元体积饱满,结构清晰,细胞内高尔基体、线粒体等趋于正常.结论抑颤汤能明显改善 PD模型大鼠的旋转行为,促进受损脑黑质细胞的修复,提示其疗效具有神经保护性特点.

Abstract:BACKGROUND:Parkinson disease(PD) has been predominately treated by Dopamine replacement at present days. Owing to its side effects and shortterm efficacy,exploration on neurological protective medication has gained more and more concentration.Since traditional Chinese medical herbs have good therapeutic efficacy and fewer side effects,study on their pharmacological mechanisms will benefit clinical practice. OBJECTIVE:To explore the effect of yichan decoction in protecting and enhancing the recovery of nigral cell of PD and its pharmacological mechanism. DESIGN:A completely randomized control study. SETTING and MATERIALS:At Experimental Animal Center of the General Hospital of Chinese PLA,SD rats were randomly divided into normal group(NG),saline group(PG) and yichan decoction group(YG). INTERVENTIONS:6-hydrodopamine was used to establish rat models with PD.Rats in YG were subjected to lavage administration with yichan decoction,and rats in NG and PG with physiological saline.From the fifth week,rat behaviors had been observed for 4 weeks in succession.All rats were decapitated as soon as medication finished,and brain slices were made for observing morphological changes of nigral cells under optical or electron microscope. MAIN OUTCOME MEASURES:Behavioral changes of rats and morphological changes of nigral cells. RESULTS:Rotational movements of rats were induced by apomorphine. The mean value of rotation for rats in NG was 22 circles/40 minutes, with no definite direction of rotation,significantly lower than 343 circles/40 minutes for rats in PD model group( t=3.390, P< 0.001) ;After treatment,it decreased to 62 circles/40 minutes in YG, statistically different from 340 circles/40 minutes in PG( t=2.648, P< 0.01) .The number of nigral cells markedly increased in YG, higher than that in PG, with clear structure of plump neuron and almost normal Golgi body and mitochondrion. CONCLUSION:Rotational behavior of PD rat can be improved by yichan decoction with the recovery of nigral cells,indicating neurological protective effect of yichan decoction.
