姜黄素联合氟尿嘧啶对结肠癌细胞增殖凋亡迁移作用

摘    要:目的:探索姜黄素调控上皮细胞间质转化(EMT)增敏5-FU的抗肿瘤作用。方法:采用姜黄素、5-FU不同给药浓度诱导结肠癌细胞株HCT8的凋亡,MTT法及克隆形成实验检测细胞增殖;划痕实验检测细胞迁移能力;Compusyn软件分析姜黄素的增敏效力;流式细胞术检测细胞凋亡情况;WesternBlot检测E-Cadherin、N-Cadherin、vimentin、cleaved-Caspase3、snail、β-catenin蛋白的表达量。结果:姜黄素能抑制HCT8细胞的增殖;6μmol/L姜黄素显著促进8μmol/L 5-FU对HCT8细胞的生长抑制和凋亡作用,联合药物指数(CI)值=0.7576;联合用药后降低HCT8细胞迁移能力,上调cleaved-Caspase3、E-Cadherin蛋白表达,同时抑制N-Cadherin、snail、β-catenin、vimentin蛋白表达。结论:姜黄素增加5-FU对HCT8细胞的敏感性,其机制可能与抑制EMT、诱导细胞凋亡有关。

Abstract:Objective:To explore the sensitizing effect of curcumin combined with 5-FU on colon cancer HCT8 cells.Methods:Curcumin and 5-FU were medicated to HCT8 cells in different methods.The proliferation of HCT8 cells was detected by MTT assay and clone formation assay.Scratch test was used to detect cell migration ability.Using Compusyn to assess curcumin synergistic effect.The effect of drug on cell apoptosis was detected by flow cytometry with Annexin V-FITC/PI staining.Western Blot was used to detect the effect of the combination on the expression of N-cadherin,E-cadherin,vimentin,cleaved-Caspase3,snail,β-catenin proteins in HCT8 cells.Results:Curcumin inhibited the proliferation of HCT8 cells.6μmol/L curcumin significantly promoted the proliferation inhibition and apoptosis of 8μmol/L 5-FU on HCT8,and the CI score valued 0.7576.The combination of the two drugs impaired cell lateral migration ability in HCT8 cells,which could up-regulate the expression of cleaved-Caspase3 and E-Cadherin protein while down-regulation of N-Cadherin,snail,β-catenin,vimentin proteins expression in HCT8 cells.Conclusion:Curcumin could increase the sensitivity of 5-FU to HCT8 cells,which might be related to EMT pathway inhibition and apoptosis.
