Effects of Compound Kushen Decoction on the protein expression of iNOS and COX-2 in colonic tissues of ulcerative colitis mice

Abstract:Objective:To observe the mechanism of Compound Kushen Decoction on inducible nitric oxidesynthase(i NOS)and cyclooxygenase 2(COX-2)proteins in the colonic mucosa of ulcerative colitis(UC)mice.Methods:Twenty BALB/c mice were randomly divided into 4 groups:normal group,model group,Compound Kushen Decoction group and mesalazine group,with 5 mice in each group.After one week of adaptive feeding,on the 8 th day,except for the normal group,the other three groups were given 3%dextran sulfate sodium(DSS)for one week to prepare a mouse model of experimental ulcerative colitis.At the same time,on the first day of modeling,the normal group and the model group were given daily gavage with distilled water,the Compound Kushen Decoction group and the mesalazine group were given daily gavage with Compound kushen Decoction and mesalazine,respectively for one week.The colonic tissues of each group were taken for pathological observation after fixation with HE staining examination.Western Blot was used to detect the expression of iNOS and COX-2 protein in colon tissue of each group.Results:Compared with the normal group,the expression levels of iNOS and COX-2 protein in the model group were significantly increased(P<0.01).Compared with the model group,the expression of iNOS and COX-2 protein in the Compound Kushen Decoction group and mesalazine group were significantly decreased(P<0.01).Conclusion:The mechanism of Compound Kushen Decoction in the treatment of ulcerative colitis may be related to the inhibition of iNOS and COX-2 protein expression in colon tissue.
