针刺调控感光细胞凋亡大鼠视网膜JAK2及STAT3表达作用机制

摘    要:目的:观察感光细胞凋亡模型大鼠视网膜JAK2和STAT3蛋白表达,探讨针刺对感光细胞凋亡引起的视网膜退行性病变的保护机制。方法:75只SD大鼠随机分为空白组、模型组、维生素A组、假针刺组和针刺组,每组15只。采用N-甲基-N-亚硝基脲(MNU)方法建立感光细胞凋亡大鼠模型。各组干预1次/d,6次/周,共4周后,HE染色和Tunel法分别观察视网膜组织形态和感光细胞凋亡结果;免疫荧光法定位和检测小胶质代表细胞-离子钙接头分子(Iba-1)活性与迁移;免疫荧光和Western Blot法检测观察蛋白酪氨酸激酶2蛋白(JAK2)和信号传导及转录激活因子3(STAT3)表达。结果:HE染色示,模型组细胞排列松散不规则,针刺组较模型组有所改善;Tunel法示,感光细胞凋亡可见针刺组凋亡率少于模型组(P<0.01);免疫荧光和Western Blot法检测结果示,针刺组Iba1、JAK2、STAT3表达显著低于模型组(P<0.01)。结论:针刺对感光细胞凋亡具有保护作用,其机制可能通过调控JAK2、STAT3蛋白抑制视网膜小胶质细胞活化与迁移有关。

Abstract:Objective:To observe the expression of JAK2 and STAT3 protein in the retina of the photoreceptor cell apoptosis model rat,and to explore the mechanism of acupuncture on the retinal degeneration caused by photoreceptor cell apoptosis.Methods:A total of 75 SD rats were randomly divided into normal control group,model group,vitamin A group,sham acupuncture group and acupuncture group,15 rats in each group.MNU(N-methyl-N-nitrosourea)method was used to establish photoreceptor cell apoptosis model rats.Intervention for each group 1 time/d,6 times/week,4 weeks later,observe the retinal morphological changes and photoreceptor cell apoptosis by HE staining and Tunel.Immunofluorescence was used to locate and detect the activity and migration of the microglial symbol Iba-1.The expression of JAK2(Janus protein tyrosine kinase 2)and STAT3(signal transducer and activator 3)were detected by Immunofluorescence and Western Blot.Results:HE staining showed the cells in the model group were loosely,disordered.The acupuncture group were improved compared with the model group.Tunel’s results of photoreceptor apoptosis showed that the number of acupuncture groups was less than that of the model group(P<0.05).Immunofluorescence detection of JAK2,STAT3 and Iba1 protein in the five groups showed that acupuncture group was significantly lower than model group(P<0.01);the Western Blot method showed that the expression of JAK2,STAT3 in the acupuncture group was lower than that in the model group(P<0.01).Conclusion:Acupuncture has protective effect on photoreceptor cell apoptosis.The mechanism may be related to the inhibition of retinal microglia activation and migration by regulating JAK2 and STAT3 proteins.
