人工虫草提取物对缺血心肌的保护作用及其机制

摘    要:目的研究人工虫草提取物对缺血心肌的保护作用及其机制。方法通过结扎大鼠左冠状动脉前降支(LAD)造成急性心肌缺血模型,观察人工虫草水提取物(Water extract from cultural mycelium,WE)、醇提取物(Ethanal extract from cul-tural mycelium,EE)、醋酸乙酯提取物(Ethyl acetate extract from cultural mycelium,EA)和石油醚提取物(Petroleum ether ex-tract from cultural mycelium,PE)对心肌梗死面积(MIS)、血清磷酸肌酸肌酶(CK-MB)、乳酸脱氢酶(LDH)、丙二醛(MDA)、一氧化氮(NO)、一氧化氮合酶(NOS)及超氧化物歧化酶(SOD)的影响。结果WE,EE和PE可明显减小MIS,降低CK-MB及LDH活性,减少MDA含量,增高NO含量,提高NOS和SOD活性。结论WE,EE和PE对缺血心肌具有保护作用,其机制与WE,EE和PE提高NOS和SOD活性从而扩张冠状动脉和抗脂质过氧化有关。

Abstract:ObjectiveTo evaluate the effects of extracts from cultural mycelium of Cordyceps sinensis on myocardial infarction and explore the possible mechanisms involved in it.MethodsRats were treated with water extract from cultural mycelium(WE),ethanal extract from cultural mycelium(EE),ethyl acetate extract from cultural mycelium(EA) and petroleum ether extract from cultural mycelium(PE)for 7 days before myocardial infarction induced by coronary occlusion.The myocardial infarct size(MIS),creatine phosphokinase(CK-MB) activity,lactate dehydrogenase(LDH) activity,nitric oxide(NO) content,malondial dehyde(MDA) content,nitric oxide synthase(NOS) and superoside dismutase(SOD) activity were measured.ResultsWE,EE and EA significantly reduced MIS,exerted notable inhibition in the elevation of CK-MB activity,LDH activity and MDA content,markedly increased NO level,NOS and SOD activities.ConclusionThe results indicated that WE,EE and EA showed a capacity to resist ischemic damage and the mechanisms were correlated with mediating NOS and SOD activity at least,in part.
