凋亡阳性率及凋亡相关蛋白Bcl-2、bax在宫颈癌放疗前后表达的意义(英文)

摘    要:
目的观察30例宫颈癌患者放疗前后的凋亡阳性率及 Bcl-2、bax 蛋白表达的变化,以探讨放射治疗肿瘤的机理。(放疗后指肿瘤组织剂量至10Gy 即 D_T=10Gy 时)。方法采用免疫组织化学方法 SP 法和脱氧核苷酸转移酶介导的末端标记技术 TUNEL 法对30例Ⅰ-Ⅲ期宫颈鳞癌患者的放疗前后凋亡阳性率及 Bcl-2、bax 蛋白表达水平进行检测。结果 30例患者凋亡阳性率放疗前后的表达分别为76.7%和100%,放疗后显著增高(P<0.05);Bcl-2蛋白在放疗前后的表达阳性率分别为73.3%和46.7%,放疗后显著降低(P<0.05);bax 蛋白在放疗前后的表达阳性率分别为86%和100%,放疗后显著增加(P<0.05)。结论放疗的主要机制是放射线诱导细胞调亡的过程,放射线可以通过抑制 Bcl-2蛋白的表达、增强 bax 蛋白的表达诱发肿瘤细胞的凋亡。

Abstract:
Objective:To evaluate the apoptosis positivity,the expression of Bcl-2.bax proteins in 30 patients with squamous cell cervix carcinoma before and after radiotherapy.Methods:By using immuno- histochemical and TDT-dUTP nick end labelling techniques,30 cases of squamous cell cervical carcinoma were analyzed.Results:The apoptosis positivity before and after irradiation was 76.7% and 100% respec- tively,with the difference being significant (P<0.05);The positive rates of Bcl-2 protein before and after irradiation were 73.3% and 46.7% respectively,with the difference being significant (P<0.05);The positive rates of bax protein be[ore and after irradiation were 86% and 100% respectively,with the difference being significant (P<0.05).Conclusion:bax and Bcl-2 protein play an important role in apoptosis induced by fractionated radiation therapy.Apoptosis induced by irradiation is contributed to upregulation of bax protein or downregulation of Bcl-2 protein.
