PGE_2对离体培养人蜕膜组织的抑制作用

取早孕妇女的蜕膜组织进行离体培养,观察PGE_2对蜕膜细胞生长的影响:蜕膜细胞在不同浓度的PGE_2的影响下皆可发生退化变性,甚至坏死。其退化变性程度随剂量增加而加剧,而正常对照组的蜕膜细胞则生长良好,实验中也对比观察芫花萜和15-甲基-PGF_(2a)的作用。按作用强度比,其顺序为芫花萜>PGE_2>15-甲基-PGF_(2a)。说明PGE_2可以直接抑制蜕膜细胞使之退化变性。此抑制作用与子宫收缩或雌、孕激素的作用或代谢无关。

THE INHIBITORY EFFECT OF PROSTAGLANDIN E_2 ON THE HUMAN DECIDUAL TISSUES CULTURED IN VITRO

The decidual tissues obtained from 12 women of early pregnancy were cultivated in vitro. The flasks with culture-medium were fivied into several groups. PGE2, 15-methyl-PGF2, and Yuanhuac-ine, all diluted at different concentration, were added into the exa-minated groups respectively.It was found that in normal coutrol groups, all the decidual cells developed very well, while in the examinated groups with PGE2. the degeneration and necrosis of the deeidual cells were observed clearly. The inhibitory effect was depended upon the concentration of PGE2. As compared the effect of PGE2 with that of 15-methyl-PGF2α and Yuanhuacine, the inhibitory potency might be arranged in such a sequance: Yuanhuacine>PGE2>PGF2. What we observsd suggests that PGE2 has the ability to inhibit the decidual cells directly; neither the contraction of the uterus nor the metabolism of estrogen or progestogen may be responsible for this effect. This inhibitory effect of PGE2 on decidual cells may be one of the mechanisms for the termination of the early pregnancy.
