涎腺肌上皮瘤中纤维形成的组织学及电镜组织化学研究

摘    要:
目的 研究涎腺肌上皮瘤细胞之间及粘液样区域中纤维的形成机制及纤维的类型。方法 采用胶原纤维及弹性纤维的组织学特殊染色方法及鞣酸-电镜组织化学技术观察4例涎腺肌上皮瘤。结果 组织学染色表明,在肌上皮瘤细胞之间及粘液样区域内分布有红染的胶原纤维(苦味酸-酸性品红法)和呈深蓝黑色的弹性纤维(间苯二酚品红法),鞣酸电镜组织化学研究结果显示,在肌上皮瘤细胞周围分布有数量不等的胶原原纤维和弹性纤维团块,在肌上皮瘤细胞的胞浆内,可见两种类型的高尔基复合体来源的分泌囊泡,一种着色深黑,电子密度均匀,内含致密的前弹性蛋白分子;另一种着色浅灰,电子密度不均匀,内含微丝状结构的前胶原蛋白分子。肌上皮瘤细胞通过胞吐作用将这些物质释放到细胞外,形成胶原纤维和弹性纤维。结论 肌上皮瘤细胞具有合成前胶原蛋白及前弹性蛋白的能力,构成肿瘤细胞外及粘液样区域中的胶原纤维和弹性纤维。

Abstract:
Objective To study the mechanism of fibrous formation and fibrous types among the tumor cells and/or in mucoid tissue of salivar myoepithelioma.Methods Four cases of salivary myoepithelioma were observed by a histological and an electron microscopic histochemical technique.Results The data of histological study showed that collagenous fibers and elastis fibers were distributed in mucoid tissue and among the tumor cells.The data of electron microscopic histochemical study showed that some slender collagen fibers and heavily stained elastic fibers were dispersed around myoepithelioma cells and in mucoid tissue.Two different types of the Golgi-derived vesicles were visualized in cytoplasm of myoepithelioma cells:Type one was small round vesicles with a uniform electron dense core of amorphous material and heavily stained;type two was small round vesicles with a non-uniform electron dense core of filamentous material and stained grey.These materials were released into the extracellular space by cytoplasmic exocytosis.Conclusion Myoepithelioma cells have the capability to synthesize and secrete procollagen and precursor of elastin,which could form collagenous fibers and elastic fibers in the stroma.
