氯沙坦钾对全氟异丁烯吸入性急性肺损伤的影响

摘    要:
目的探讨肾素-血管紧张素系统(renin—angiotensinsystem,RAS)中血管紧张素1I(angiotensinll,Angll)在全氟异丁烯(perfluoroisobutylene,PFIB)吸入性急性肺损伤(acutelunginjury,ALI)中的作用。方法使用全身暴露PFIB染毒装置对CDl小鼠实施染毒,采用Anglll型受体(anglll-typereceptor,ATl)特异性抑制剂氯沙坦钾作为工具药,以肺系数、支气管肺泡灌洗液(bronchoalveolarlavagefluid,BALF)总蛋白含量及72h内存活率为观察指标,评估ATl受抑制状态下肺损伤情况。结果与结论在PFIB染毒前0.5h、染毒后0.5h、1h经腹腔注射方式给予实验CDl小鼠氯沙坦钾(40mg/kg),虽然对各项ALI指标均有一定的改善趋势,但并不能起到显著的防治效果。以AngII为主要成分的RAS可能并不参与PFIB吸入性急性肺损伤早期炎症反应的扩大过程。

Abstract:
Objective To investigate the therapeutic effect of angiotensin 11 on PFIB-inhalation induced acute lung inju- ry(ALI). Methods CD1 mice were exposed to PFIB under the systemic exposure device and treated with losartan potassi um, an AT1 specific inhibitor, at the predetermined time. To estimate the degree of ALI when AT1 was almost blocked, the total protein content in BALF and lung coefficients were detected. Meanwhile, the survival ratio during 72 hours after PFIB exposure was counted. Results and Conclusion The tests showed that the lung coefficients and total protein content in BALF were slightly attenuated after the treatment with losartan potassium at a dose 40 mg/kg, but were of no statistical signif icance between the medicated groups. Losartdn potassium seemed to have no therapeutic effect on PFIB poisoning. The results suggested that RAS might not participate in the excessive inflammation during the process of PFIB-inhalation induced ALL
