联胺诱导下丘脑-垂体对MDA代谢、ICAM-1表达及动脉粥样硬化形成的影响

摘    要:目的 探讨离体培养下丘脑-垂体受联胺诱导前后对内皮细胞丙二醛(Malondialdehyde,MDA)代谢、细胞间粘附分子-1(Intercellular Adhesion Molecule-1,ICAM-1)表达的作用以及对动脉粥样硬化(Atherosclerosis,AS)形成的影响。方法 将培养内皮细胞随机分为两组:正常组及受损组。每组又再分为3小组:HP组、dHP组、Control组(对照组)。分别培养于预先收集的下丘脑-垂体(HP组)、联胺诱导下丘脑-垂体(dHP组)和对照组(Control组)条件培养液。检测各组上清MDA含量;免疫细胞化学方法观察内皮细胞ICAM-1的表达。结果 联胺作用后内皮细胞MDA含量明显升高,ICAM-1相对高表达。各组下丘脑-垂体对内皮细胞都有下调MDA含量及ICAM-1表达的作用。联胺诱导后下丘脑-垂体对正常内皮细胞的下调作用更大;对受损内皮细胞这种下调作用却明显减弱。结论 联胺诱导后离体下丘脑-垂体对正常内皮细胞起抗损伤作用。可抑制AS形成;对受损内皮细胞的抗损伤作用减弱,可促进AS形成。

Abstract:objective To observe the changes of MDA metabolism and ICAM-1 expression of endothelial cells affected by the hypothalamus-pituitary which had been induced by diamide in vitro and to investigate the influence of hypothalamic vessels' center on pathogenesis of AS.Methods The cultured endothelial cells were randomly divided to two group: normal EC and damaged EC. Each group was cultured separately with various conditioned media collected respectively as hypothalamus-pituitary(HP),hypothalamus-pituitary induced by diamide(dHP),and control. Then the further cultured supernatants were collected respectively for detecting the content of MDA and the cells were fixed for immunocytochemical staining to detect the expression of ICAM-1.Results Diamide up)-regulated the metabolism of MDA and expression of ICAM-1 of EC.The hypothalamus-pituitary of all experimental groups showed a down regulation in the metabolism of MDA and expression of ICAM-1 of normal EC,especially in the dHP group;while the down-regulation effect showed significant depression on dHP group of the endothelial cells induced by diamide ascompared with those of HP group.Conclusion The hypothalamus-pituitary induced by diamide shows anti-damage effects on the normal EC and failure of anti-damage effects on damaged EC which may be related to the pathogenesis of atherosclerosis.
