胸腺肽乙基纤维素微囊的制备和体外释药研究

摘    要: 目的为提高胸腺肽的生物利用度,增强疗效,制备了胸腺肽乙基纤维素缓释微囊。方法用液中干燥法制备胸腺肽乙基纤维素微囊,正交设计法筛选其最佳制备工艺,Lowry法测定药物的含量,计算微囊的载药量、包封率及体外释药量。结果微囊粒径范围为30-80ym,平均粒径为52.64 tim,平均含药量为20.420h,平均包封率为81.64%,其体外释药符合Higuchi方程Q=5.15387+35. 350 59t1/2(r=0.997,n=9),f1/2-80min,稳定性考察实验结果表明其稳定性较好。结论本法制备的胸腺肽乙基纤维素微囊粒径分布集中,体外释药有明显的缓释作用,具有良好应用前景。

Abstract:OBJECTIVE To study the preparation of thymopeptide ethylcellulose sustained-release microcapsule for improving the bioavailability and curative effect of thymopeptide.METHODS Intra-liquid desiccation method was used to prepare thymopeptide ethylcellulose microcapsule, and orthogonal design was used to optimize the technology of preparation. The drug content was exam-ined by Lowry methods, and the drug loading, encapsulation efficiency, and drug release in vitro were calculated.RESULTS The diameter of the THT-EMC was in the range of 30-80Um, and the average particle diameter was 52.64 pnl.The average drug con-tent and the average encapsulation efficiency were 20.42%and 81.64%respectively. The drug release in vitro can be described by model of Higuchl equation as Q= 5.153 87+ 35 .350 59tli-(r=0.997,n=9), with the tl/2 in vitro was 80 min. Almost no changes were observed in size distribution ,drug content, and drug release after the THT-EMC were stored at the temperature of 40cor the illumination of 12 000-4 000 I_X.CONCIJUSION The size distribution of THT-EMC was focalizing. It was found that THT-EMC had obviously sustained effect in vitro and favorable application foreground.
