糖尿病对大鼠阴茎组织中eNOS和nNOS的影响及胰岛素的治疗作用

摘    要:目的:研究糖尿病(DM)对大鼠阴茎组织中内皮型一氧化氮合酶(eNOS)和神经型一氧化氮合酶(nNOS)蛋白表达的影响及胰岛素的治疗作用。方法:随机选取雄性SD大鼠70只,其中60只以链脲佐菌素(STZ)诱导建立DM模型后,行阿朴吗啡阴茎勃起实验,筛选DM性ED模型。以未加处理因素的10只大鼠作为正常对照组,发生ED者随机分为DM性ED组、胰岛素组,发生DM但未发生ED者作为DM组,未成DM者作为STZ组。各组干预6周后,采用免疫组织化学SP法检测各组大鼠阴茎组织中eNOS、nNOS蛋白表达。结果:DM组、DM性ED组大鼠阴茎组织中eNOS、nNOS蛋白表达水平显著低于正常对照组(P<0.01);胰岛素组显著高于DM组和DM性ED组(P<0.01),其中,eNOS蛋白表达水平接近于正常对照组(P>0.05),nNOS蛋白表达水平低于正常对照组(P<0.01);STZ组与正常对照组之间eNOS、nNOS蛋白表达差异均无统计学意义(P>0.05)。结论:(1)DM可以显著降低大鼠阴茎组织中eNOS、nNOS蛋白表达水平,提示其可能是DM性ED发病的重要机制之一;(2)胰岛素干预可以减缓DM性ED大鼠阴茎组织中eNOS、nNOS蛋白表达的下降。

The study on eNOS and nNOS expression in the corpus cavernosum tissue of diabetic rats
Abstract:Objective: To study the eNOS and nNOS expression in the corpus cavernosum tissue of diabetic rat and the therapeutic effects of insulin. Methods: Sixty male Sprague-Dwaley rats with normal erectile function were injected with streptozotozin to make experimental models of diabetes melliuts (DM), then injected with apomorphine in all of the DM rats to selecte erectile dysfunction rats models with dibaetes. There were 10 nomral rats in the normal group, the ED rats were devided into the DM with ED group, the insulin group, the diabetic rats without ED were called the DM group; the rats did not with diabetes were called the STZ group. The eNOS and nNOS expression were detected by means of SP immunohistochemistry. Results: The eNOS, nNOS expressions in DM with ED group and DM group were significantly lower than those in control group (P<0.01); those in the insulin group were higher than DM with ED group (P<0.01) and DM group (P<0.01), and the eNOS protein level was closer to the normal control group (P>0.05), nNOS was lower than that in control group (P<0.01); The eNOS, nNOS expression in STZ group compared with control group had no significant difference (P>0.05). Conclusion: (1) Diabetes can reduce the eNOS and nNOS expression in penile tissue of rats, it suggested that diabetes may be one of the important pathogenesis of DM with ED;(2)Insulin intervention can inhibit eNOS, nNOS contents.
