青蒿琥酯抑制人肝癌细胞株HepG2细胞生长及其机制的初步研究

摘    要:目的:研究青蒿琥酯对人肝癌细胞株HepG2细胞增殖和survivin表达的影响,探讨青蒿琥酯影响肝癌细胞增殖的可能机制.方法:常规培养人肝癌细胞株HepG2,设立空白对照组;青蒿琥酯不同浓度组(3.125μg/ml、6.25μg/ml、12.5μg/ml、50μg/ml),作用不同时间后用MTT法检测青蒿琥酯对人肝癌细胞株HepG2细胞增殖的影响,应用RT-PCR检测细胞survivin在mRNA水平的表达.结果:青蒿琥酯作用人肝癌细胞株HepG2后,与对照组比较细胞生长明显受到抑制(P〈0.05),当青蒿琥酯浓度在12.5μg/ml时,survivin的表达(相对表达量为(0.75+0.01)显著降低(P〈0.05).结论:青蒿琥酯可通过下调survivin的表达来抑制肝癌细胞HepG2的生长.

Primary research on artesunate inhibiting human hepatoma cell lines HepG2 cells growth and its mechanisms

Abstract:Objective:Its aimed to investigate the effects of artesunate on proliferation and surviving expressions of human hepatoma cell lines HepG2,human hepatoma cell lines HepG2 cultivated by convention, were randomly divided into 2 groups:untreated control group; different concentration artesunate treated with HepG2. And then, the proliferation of HepG2 were detected by MTT; After treated with different concentration for 48 hours,using RT-PCR method to detect survivin expression at the mRNA level. Results:Compared with the control group, the proliferation of HepG2 was significantly inhibited by artesunate(P 〈 0.05) ;when the concentration of artesunate reach to 12.5μg/ml, the survivin expressionin(relative expression levels of 0. 75±0. 01) were significantly lower(P 〈 0.05). Conclusion: Artesunate can inhibited the proliferation of H epG2 through reducing the expressionin of the surviving.
