摘    要:
目的 探讨新型趋化因子CKLFSF8对人白血病细胞株增殖的影响。方法 RT—PCR检测CKLFSF8基因在细胞株K562中的表达;利用脂质体将CKLFSF8转染K562;倒置显微镜观察细胞生长,MTT法检测CKLFSF8基因转染前后肿瘤细胞增殖的变化。结果 细胞生长过程中可测到CKLFSF8表达;CKLFSF8转染对K562的增殖有抑制作用,与空白对照组和空质粒组有显著性差异(P〈0.05);结论 CKLFSF8基因转染对人白血病细胞株K562的增殖有明显抑制作用,可望在肿瘤治疗中发挥重要作用。

Abstract:
Objective To investigate the effect of a novel chemokine CKLFSF8 on cell proliferation of tumor cells. Methods Expression of CKLFSF8 mRNA on K562 cell line was assayed by RT-PCR.The target gene was transfected into the cell in the way of lipid,and the cell growth was observed by inverted microscope.Cell proliferation was determined by MTT assay. Results CKLFSF8 was expressed in K562.After transfection,cell proliferation was inhibited(P<0.05). Conclusion The result shows that cell proliferation can be inhibited by transfection of CKLFSF8.The novel chemokine may have a bright prospect in tumor therapy.
