康莱特注射液对Lewis肺癌小鼠增殖细胞核抗原表达的影响

摘    要:目的通过检测增殖细胞核抗原(PCNA)的表达,阐明康莱特注射液的抑瘤作用及其可能的作用机制。方法50只C57小鼠复制Lewis肺癌模型后,随机分为5组。分别腹腔注射相应药物14 d后处死,称瘤质量,计算抑瘤率,免疫组化法检测各组小鼠肺癌组织中PCNA蛋白表达。结果CTX组、康莱特低剂量组、康莱特中剂量组、康莱特高剂量组抑瘤率分别为60.71%,45.24%,47.02%,55.36%;PCNA的阳性表达率分别为(90.78±5.51)%,(53.63±3.56)%,(72.87±4.12)%,(67.61±3.23)%,(53.51±3.20)%。结论抑制PCNA在肿瘤细胞中的表达可能是康莱特注射液抑制肿瘤生长的机制之一。

Abstract:Objective It is to interpret the restraining tumor action of Kanglaite injection and the possible mechanism of action through detecting the expression of proliferating cell nuclear antigen(PCNA).Methods 50 C57 mice copied Lewis lung cancer model were randomly divided into five groups.The mice were sacrificed after injecting corresponding drugs with intraperitoneal injection for 14 d.The mass of tumor was weighed and the rate of restraining tumor was calculated.The expressions of PCNA protein in the tissue of lung cancer in all the groups were detected with immunohistochemical method.Results The rate of restraining tumor in CTX group,Kanglaite low dosage group,Kanglaite middle dosage group and Kanglaite high dosage group was 60.71%,45.24%,47.02% and 55.36% respectively.The positive expression rate of PCNA in model group,CTX group,Kanglaite low dosage group,Kanglaite middle dosage group and Kanglaite high dosage group was(90.78±5.51) %,(53.63±3.56) %,(72.87±4.12) %,(67.61±3.23) % and(53.51±3.20) % respectively.Conclusion Restraining the expression of PCNA in tumor cell maybe one of the mechanism of Kanglaite injection restraining the growth of tumor.
