泰山磐石散对流产模型大鼠母胎界面FAS、FASL表达的影响

摘    要:
目的:研究泰山磐石散对流产模型大鼠母胎界面FAS、FASL表达的影响,从而探讨泰山磐石散保胎的作用机制。方法:将雌性 SD 大鼠和雄性 SD 大鼠按1:1的比例合笼喂养,然后第二天观察是否有阴栓,若观察到有阴栓则标记为妊娠的第一天。将全部受孕大鼠阴栓随机分3组,分别为正常组,模型组,治疗组。正常组在怀孕后6~8d每天进行皮下注射75%乙醇溶液(75%乙醇为溴隐亭的溶解剂),而在怀孕后的1~11d每天灌服蒸馏水;模型组在怀孕的6~8d则每天皮下注射溴隐亭溶液,在怀孕的1~11d灌服蒸馏水;治疗组在怀孕后6~8d皮下注射溴隐亭溶液,在怀孕的1~11d灌服泰山磐石散。第12天取蜕膜组织利用免疫组化法检测母胎界面子宫侧蜕膜组织FAS、FASL表达情况。结果:与正常组相比,模型组母胎界面子宫侧蜕膜组织FAS表达均上升,FASL表达均下降(P<0.01);模型组相比,治疗组蜕膜组织母胎界面子宫侧FAS表达均下降,FASL表达均上升(P<0.01)结论:泰山磐石散治疗复发性自然流产一方面是通过下调蜕膜组织母胎界面中FAS表达水平,另一方面是上调FASL表达水平,从而来达到减少蜕膜组织中细胞凋亡的目的,最终达到其降低流产率的效果。

Abstract:
Objective:To study the effect of Taishan Panshi Powder on the expression of FAS and FASL in the maternal-fetal interface of abortion model rats,and to explore the mechanism of Taishan Panshi Powder to protect the fetus.Methods:Female SD rats and male SD rats were fed in a cage at a ratio of 1:1,and then the next day was observed for the presence of a penis,and if it was observed,it was marked as the first day of pregnancy.The vaginal plugs of all pregnant rats were randomly divided into 3 groups:normal group,model group and treatment group.The normal group received a subcutaneous injection of 75% ethanol solution (75% ethanol is bromocriptine’s dissolving agent) 6 to 8 days after pregnancy,and distilled water was administered daily from 1 to 11 days after pregnancy.The model group was administered 6 to 8 days after pregnancy.The bromocriptine solution was injected subcutaneously every day,and distilled water was administered on the 1st to 11th days of pregnancy.The treatment group was subcutaneously injected with bromocriptine solution on the 6th and 8th days after pregnancy,and the Taishan Panshi powder was administered on the 1st to 11th days of pregnancy.On the 12th day,the decidual tissue was collected and the expression of FAS and FASL in the uterine decidual tissue at the mother-fetal interface was detected by immunohistochemistry.Results:Compared with the normal group,the expression of FAS in the uterine decidual tissue of the maternal-fetal interface of the model group increased,and the expression of FASL decreased (P<0.01).Compared with the model group,the expression of the FAS of the uterine side of the decidual tissue of the decidual tissue of the treatment group decreased.The expression of FASL increased (P<0.01).Conclusion:Taishan Panshi powder treatment of recurrent spontaneous abortion is to reduce the expression of FAS in the maternal-fetal interface of decidual tissue on the one hand,and to increase the expression of FASL on the other hand to reduce decidua The purpose of cell apoptosis in the tissues finally achieves its effect of reducing abortion rate.
