NO、SOD在糖尿病大鼠早期肾损害中的作用

摘    要:目的 :探讨一氧化氮 (NO)及超氧化物歧化酶 (SOD)在糖尿病 (DM)早期肾损害中的作用。方法 :将实验鼠分为正常对照组 (A组 )、糖尿病组 (B组 )、糖尿病 + 0 .5 %左旋 精氨酸 (L Arginine)组 (C组 )。检测各组第 1,2周内生肌酐清除率 (Ccr)、尿微量白蛋白排泄率 (UAE)、尿亚硝酸盐 /硝酸盐 (NO-2 /NO-3 )、肾皮质NO-2 /NO-3 和总 超氧化物歧化酶 (T SOD)、铜锌 超氧化物歧化酶 (CuZn SOD)、肾重 /体重及肾小球平均体积 (AGV)。结果 :第 1,2周B、C组Ccr、UAE、尿NO-2 /NO-3 较A组明显升高 (P <0 .0 1) ,C组Ccr、UAE、尿NO-2 /NO-3 较B组显著升高 (P <0 .0 5 ) ,第2周B、C组肾皮质NO-2 /NO-3 较A组显著升高 (P <0 .0 5 ,0 .0 1) ,C组较B组显著升高 (P <0 .0 5 ) ,B、C组T SOD、CuZn SOD较A组显著下降 (P <0 .0 1) ,C组较B组更低 (P <0 .0 5 )。第 2周C组肾重 /体重、肾小球平均体积较B组显著升高。结论 :糖尿病肾病早期肾脏抗氧化酶活性减弱 ,超氧阴离子增多 ,内源性NO合成增加 ,补充L Arginine加重糖尿病肾损害。

Abstract:Objective:To investigate the effects of nitric oxide(NO)and superoxide dismutase (SOD) on early renal injury of diabetic rats.Methods:Rats were diveded into normal control group (A),diabetic rat group (B) and diabetic 0.5% L-arginine group (C).Ccr,UAE,urinary NO - 2/NO - 3,renal cortx NO - 2/NO - 3,T-SOD,CuZn SOD,rats kidney/body weight and average glomerular volume (AGV) were measured in the first and second week.Results:At the beginning of the first week,Ccr,UAE and urinary NO - 2/NO - 3 increased significantly in groups B and C compared with those in group A (P<0.05) and increased significantly in group C compared with those in group B(P<0.05).Renal cortex NO - 2/NO - 3,kidney/body weight and AGV increased significantly in groups B and C compared with those in group A and increased significantly in group C compared with those in group B.But renal cortex T-SOD and CuZn-SOD decreased more significant in groups B and C than those in group A.Conclusion:The activities of T-SOD and CuZn-SOD weakened,but NO increased in early diabetic nephropathy.L-arginine can increase the early renal injury of diabetic rats.
