| 摘 要： | 目的 检测脂质体介导增殖细胞核抗原 (PCNA)反义寡聚核苷酸抑制子宫颈癌细胞体外增殖活性效果。方法 应用细胞生长曲线和MTT比色法检测细胞增殖活性 ,并采用免疫组织化学方法 (SABC法 )检测PCNA蛋白的表达。结果 反义寡聚核苷酸处理组细胞与对照组比较生长曲线差异有显著性 ,增殖显著受抑 (P <0 0 5 ) ,PCNA蛋白表达完全抑制。而正义寡聚核苷酸组则无此作用 (P >0 0 5 )。结论 提示PCNA反义寡聚核苷酸可以显著抑制子宫颈癌细胞体外增殖活性 ,其抑制作用可能通过阻断PCNA蛋白表达实现

| Abstract: | Objective To evaluate the inhibitive effect of liposome conjugated antisense oligonucleotides on uterine cervix carcinoma cell line proliferation. Methods HeLa cell line was treated with antisense oligodeoxynucleotides (ASODN) targeting the mRNA encoding proliferating cell nuclear antigen (PCNA). The inhibition of proliferation was studied by cell growth curves and MTT method. The expression of PCNA was detected immunohistochemically. Results The cell proliferation was inhibited effectively by ASODN ( P <0 05). The protein expression of PCNA was inhibited completely. The sense oligodeoxynucleotides (SODN) showed no such effect ( P >0 05). Conclusion Antisense oligonucleotides can obviously inhibit the proliferation of uterine cervix carcinoma cell by inhibiting the expression of PCNA. |
