山茱萸总苷对坐骨神经损伤 Balb/c 小鼠神经传导速度及脊髓运动神经元内 Gap-43表达的影响

摘    要:
目的:探讨山茱萸总苷促进坐骨神经损伤再生及修复的临床效果及机制。方法84只 Balb/c 小鼠随机分为山茱萸总苷高剂量组(320 mg/kg/d),中剂量组(160 mg/kg/d),低剂量组(80 mg/kg/d)及生理盐水对照组,每组16只。各组分别于术后3天、5天、1周、2周、4周、8周时间点采用免疫组化法检测 L4~ L6脊髓运动神经元内生长相关蛋白(Gap-43)表达;术后第4、8、12周行电生理检查测定神经传导速度(MCV)。结果高剂量组,中剂量组 Gap-43表达明显多于低剂量和对照组(P <0.05),术后第4、8、12周高剂量组、中剂量组较低剂量组和对照组再生神经传导速度加快、波幅升高、潜伏时缩短(P <0.05)。结论山茱萸总苷能够促进大鼠周围神经损伤后 MCV 的恢复,其机制可能为增加损伤脊髓运动神经元中 Gap-43的表达。

Abstract:
Objective To investigate the clinical effect of cornus officinalis glycosides(COG)and its mechanism to promote the regeneration of sciatic nerve injury.Methods 84 Balb/c mice were randomly divided into COG high dose group(320mg/kg/d),middle dose group (160 mg/kg/d),low dose group (80 mg/kg/d)and physiological saline control group,with 16 mice in each group.The sciatic nerve of the four groups were observed by immunohistochemistry at 3,5 day and 2,4,8,12 weeks after operation to study the expression of Gap-43 of L4~L6 motoneurons and the nerve con-duction velocity (MCV)of electrophysiological examination at 4,8,12 weeks.Results The expression of Gap-43 of high dose group and middle dose group were higher than the low dose and the control group at 3,5 day and 2,4,8,12 weeks (P 〈0.05);after fourth,8,12 weeks of high dose group,middle dose group and low dose group than in the control to shorten the regeneration of nerve conduction speed,increase the amplitude,latency (P 〈0.05).The regenerated nerve of the COG groups had faster conduction velocity,higher amplitude and shorter latency time than the control group at 4,8,12 weeks after the operation.Conclusion COG can promote the recovery of MCV,the mechanism may up-regulate the Gap-43 expression of the motoneurons in the damage spinal cord.
