原花青素对2型糖尿病局灶性脑缺血大鼠海马组织兴奋性氨基酸的影响

摘    要:目的探讨原花青素(PC)对2型糖尿病局灶性脑缺血大鼠海马组织兴奋性氨基酸(EAA)的影响。方法通过高脂高糖饮食4 wk,尾静脉注射小剂量链脲佐菌素(STZ)25.0 mg·kg~(-1)制备2型糖尿病大鼠模型,2 wk后建立局灶性脑缺血模型,24 h后处死,高效液相色谱法测模型大鼠海马组织EAA,观察PC(50、100、200 mg·kg~(-1))对EAA的影响。结果与假手术组比较,普通脑缺血大鼠模型和2型糖尿病缺血模型谷氨酸(Glu)和天冬氨酸(ASP)及抑制性氨基酸γ-氨基丁酸(CABA)均有升高(P<0.05,P<0.01),但2组间无明显差异(P>0.05),对甘氨酸(Gly)无明显影响(P>0.05);与2型糖尿病缺血模型组相比,PC中、高剂量组Glu、ASP显著下降(P<0.01),各剂量组对GABA和Gly无明显影响。结论PC可能通过对抗EAA毒性实现对2型糖尿病局灶性脑缺血大鼠的保护作用。

Abstract:AIM To investigate the effect of procyanidin (PC) on the excitatory amino acids (EAA) of hippocampus in type 2 diabetes mellilus (T_2DM) rats with focal cerebral ischenaia.METHODS T_2DM rats were made by feeding fat and glucose forage for 4 wk,and injecting streptozotocin (STZ) through caudal vein in low dose (25.0 mg·kg~(-1)).Two weeks later,the focal cerebral ischemia modes were made and rats were killed 24 h later,and the effect of PC (50,100,200 mg·kg~(-1)) on EAA in hippocampus was observed by high performance liquid chromatograph.RESULTS Compared with sham-operation group,the glutamic acid (Glu), aminosuccinic acid (ASP) and gamnaa-anaionbutyric acid (GABA) increased in comnaon and T_2DM model rats (P<0.05,P<0.01) with no difference for the two groups (P>0.05),and there were no difference in glycine (Gly) (P>0.05) in the two groups.Compared with T_2DM model group,Glu and ASP of mid-dose and high-dose groups decreased (P<0.01,P<0.05),and PC had no effect on GABA and Gly in all dose groups (P>0.05).CONCLUSION By resisting to the toxicity of EAA,PC may realize the neuroprotective effect on ischemic rats of type 2 diabetes mellitus.
