武当青牛胆化学成分与抑菌活性的初步研究

摘    要:
 目的 对武当青牛胆[Tinospora sagittata (Oliv.) Gagnep.]的化学成分进行研究,并初步筛选具有抑菌活性的化合物。方法 对青牛胆干燥块根进行渗漉提取,用柱色谱方法(Silica gel, MCI, Rp-8等)对乙酸乙酯部位提取物进行分离纯化,并根据理化性质和光谱数据(MS,1H-NMR,13C-NMR)进行结构鉴定。单体化合物采用比浊法测定最小抑菌浓度(MIC)。结果 从武当青牛胆中分离得到5个化合物,分别鉴定为古伦宾、异古伦宾、药根碱、金果榄苷、巴马汀,其中巴马汀抑菌活性最强,药根碱次之,古伦宾、异古伦宾和金果榄苷最弱。结论 本实验首次以武当道地药材青牛胆为材料进行化学成分分析和单体活性研究,为武当青牛胆的活性成分研究开发提供了理论依据,对于阐明其药理作用的物质基础具有一定的指导意义。

Abstract:
OBJECTIVE To study chemical constituents and antimicrobial effect of Tinosporae sagittata (Oliv.) Gagnep. METHODS The compounds were isolated from ethanol extraction and purified by MCI, Rp-8, silica gel column chromatograph. The structures of the compounds were determined by physicochemical properties and the spectrum analysis (TLC, MS, 1H-NMR, 13C-NMR). The antimicrobial abilities of the five compounds were determined by colorimetry. RESULTS Five compounds were separated from Tinospora sagittata and were identified as columbin (Ⅰ), isocolumbin (Ⅱ), jatrorrhizine (Ⅲ), tinoside (Ⅳ) and palmatine (Ⅴ). The antimicrobial activity of palmatine was the strongest, jatrorrhizine was relatively weak. CONCLUSION The compound Ⅰ-Ⅴ all were isolated from WuDang Tinosporae sagittata (Oliv.) Gagnep. for the first time.
