消化道恶性肿瘤患者纤维蛋白胶包埋顺铂腹腔给药的药动学

 目的 研究纤维蛋白胶包埋顺铂缓释化疗的药动学特点。方法 将38例消化道恶性肿瘤患者随机分为2组： 纤维蛋白胶包埋顺铂化疗20例为试验组，普通顺铂腹腔化疗18例为对照组。测定并比较两组各个时间点的血、尿顺铂浓度，计算其半衰期(t1/2)、表观分布容积(apparent volume of distribution ,V_d)和曲线下面积(area under the curve, AUC)。结果 两组患者化疗后的血和尿浓度、t1/2、V_d及AUC差异均有统计学意义(P<0.01)。结论 纤维蛋白胶包埋顺铂缓释化疗的t1/2较普通腹腔化疗长，药物主要分布在腹腔局部，提高腹腔局部化疗药物的浓度。

Pharmacokinetic Study of Cisplatin Combined in Fibrin Glue Intraperitoneal Chemotherapy in Patients with Gastrointestinal Cancer

OBJECTIVE To study the cisplatin combined in fibrin glue intraperitoneal chemotherapy in patients with gastrointestinal cancer. METHODS 38 cases with gastrointestinal cancer were divided into two groups. Cisplatin combined in fibrin glue intraperitoneal chemotherapy were implanted in 20 patients undergoing gastrointestinal cancer surgery in the experimental group, while cisplatin were implanted in 18 cases undergoing gastrointestinal cancer surgery served as controls. The cisplatin drug concentration in plasma and urine, half-life (t1/2), apparent volume of distribution(V_d), area under curve(AUC) were compared among the two groups. RESULTS Cisplatin combined in fibrin glue intraperitoneal chemotherapy had high apparent volume of distribution, long half life and small area under the curve compared with controls. The differences were statistically significant (P<0.01). CONCLUSION Cisplatin combined in fibrin glue intraperitoneal chemotherapy can prolong the regional action time and improve the regional concentration of chemotherapeutic drugs.
