摘    要:
目的 :探讨微量元素锌对微波致视网膜神经节细胞的脂质过氧化损伤的防护作用 ,为进一步研究微波损伤机理及其防护提供一定的实验依据。方法 :体外培养猪视网膜神经节细胞 ,按微波辐照时间分为对照组、3 0 min组、6 0 min组 (辐照强度为 3 0 m W· cm-2 ) ,以及各辐照时间加锌组 ,2 4 5 0 MHz微波理疗机于微波屏蔽室内辐照 1 h,辐照后观察细胞形态 ,收集细胞 ,测定超氧化物歧化酶 (SOD)和丙二醛 (MDA)含量 ,测定细胞内锌含量。结果 :微波辐照后 ,细胞有聚集现象 ,部分细胞轴突消失 ,细胞 MDA含量明显增高 ,SOD降低 ,细胞内锌含量降低 ;加锌各组光镜下细胞形态变化不明显 ,MDA含量有所恢复 ,SOD含量增高 ,细胞内锌含量显著增高。结论 :微波可引起视网膜神经节细胞脂质过氧化损伤 ,锌可提高细胞的抗氧化能力 ,在一定程度上减轻微波对视网膜神经节细胞的过氧化损伤

Abstract:
AIM:To determine the protection of zinc on lipid peroxidation damage in the primary cultured pig retinal ganglion cells induced by microwave can provide some experiment foundation for study of mechanism of microwave damage and its protection.METHODS:Pig retinal ganglion cells were cultured in vitro and divided into 3 groups of different radiation times (control group,30 min group,60 min group,with radiation intensities of 30 mW·cm -2 )and 2 more radiated groups with zinc added.The morphological variation of cells was observed immediately after radiation.The activity of intracellular superoxide dismutase(SOD),the content of malondialdehyde(MDA)and zinc of cells were assayed after they had been radiated for 1 h in a shielded room by 2450MHz continuous wave physiotherapy machine.RESULTS:After radiation,some cells congregated and their axon disappeared.The content of MDA was increased and SOD and zinc decreased clearly compared with control group.Cells had not obvious changes observed with optics microscope in zinc added groups.content of MDA recovered,and SOD increased.The content of zinc was increased in zinc added groups.CONCLUSION:The results of the study indicated that microwave can induce lipid peroxide damage to in vitro cultured retinal ganglion cells and that zinc can improve antioxidativ capacity of cells,and to some extent can remedy microwaveinduced damage of lipid peroxidation to retina ganglion cells.
