| 摘 要： | 目的 观察严重烧伤早期小鼠腹腔巨噬细胞c-Jun激活区结合蛋白1(JAB1)蛋白表达变化,探讨安定-氯胺酮对其表达变化的影响.方法 将成年健康雄性昆明小鼠随机分为4组,即对照组、严重烧伤组、安定-氯胺酮组和严重烧伤后安定-氯胺酮处理组,以20%～30%体表总面积(TBSA)Ⅲ度烧伤作为严重烧伤模型,肌肉注射安定0.4mg/kg、氯胺酮10mg/kg给予麻醉.收集腹腔巨噬细胞提取总蛋白,运用Western blotting方法检测各组小鼠腹腔巨噬细胞JAB1蛋白表达水平的变化.结果 在严重烧伤后30min,小鼠腹腔巨噬细胞JAB1蛋白表达水平与对照组相比明显降低(P<0.05),于6h时达到最低(P<0.01),严重烧伤后12h逐渐恢复至正常水平.然而,严重烧伤后应用安定-氯胺酮麻醉,小鼠腹腔巨噬细胞JAB1蛋白表达水平在各时相点变化差异无统计学意义.结论 严重烧伤可引起小鼠腹腔巨噬细胞JAB1蛋白表达水平的早期下降,而严重烧伤后应用安定-氯胺酮麻醉能显著抑制JAB1蛋白表达水平下调.
