人参皂甙对应激大鼠肝脏热休克蛋白70表达的影响

摘    要:
目的研究人参皂甙对应激时肝脏热休克蛋白(HSP)70表达的影响,探讨其抗应激的作用机制。方法54只SD大鼠分为三组:干预组和模型组各24只,分别以人参皂甙溶液或生理盐水灌胃;对照组6只,予正常饮食。采用盲肠结扎穿孔法(cecumligationandperforation,CLP)制作应激模型。分别于造模后6、12、24及72h检测大鼠肝细胞HSP70的表达及血清IL-6水平的变化。结果除12h外,干预组肝细胞HSP70的表达均〉对照组(P〈0.05)。干预组72h肝细胞HSP70表达〉模型组(P〈0.05)。干预组血清IL-6水平整体呈下降趋势,除6h外,各时点明显〈模型组(P〈0.05)。结论人参皂甙可提高应激时大鼠肝细胞HSP70的表达,并可抑制IL-6的产生,减轻炎症反应,保护应激时的机体。

Abstract:
Objective To study the effect and mechanism of panaxoside on the expression of liver heat-shock protein-70 in condition of stress.Methods 54 SD rats were divided into panaxoside treating group(n=24),which were fed with 0.5% panaxoside liquor at 50mg/kg via gastric tube,stress model group(n=24)and normal control group(n=6).Cecum ligature and perforation(CLP)method were used to establish the model.The expression of HSP-70 and serum concentration of IL-6 were evaluated at 6h,12h,24h and 72h after operation.Results In panaxoside treating group,the expression of HSP-70 was higher(p〈0.05)than normal group at every time point except 12h,and in stress model group it is higher(p〈0.05)than in normal group at 72h after operation.IL-6 level were decreased after CLP in panaxoside group,and it was lower in panaxoside group than in stress model group 12h、72h after CLP(p〈0.05).Conclusions Panaxoside can promote the expression of rat liver cell HSP-70 so as to protect body under stress and inhibit the production of IL-6 in some way and release the inflammatory reaction.
