重组人脑利钠肽对兔髂动脉球囊损伤后血管重构及VEGF阳性表达的影响

摘    要:目的观察重组人脑利钠肽对球囊损伤的动脉血管壁血管重构及VEGF阳性表达的影响。方法随机将20只雄性新西兰大白兔分为2组,构建兔髂动脉球囊损伤模型。术后对照组只喂标准兔饲料,实验组喂标准兔饲料及重组人脑利钠肽0.1 mg·kg~(-1)·d~(-1)皮下注射20 d。术后4周,活体取损伤的动脉组织。制作常规病理切片以及VEGF免疫组化切片。采用病理图像分析系统测定血管重构的各项指标。结果实验组血管内弹力膜下面积、外弹力膜下面积、残余管腔面积较对照组增大(P〈0.05,P〈0.01),面积狭窄率较对照组缩小(P〈0.01),2组中膜面积无显著性差异(P〉0.05)。实验组较对照组内皮细胞浆、平滑肌细胞浆内VEGF表达减弱。结论重组人脑利钠肽可改善兔髂动脉球囊损伤后血管重构,抑制VEGF阳性表达。

Abstract:Objective To reveal the influence of nesiritide on vascular remodeling and VEGF expression in rabbits after angioplasty in iliac artery.Methods Twenty male New Zealand rabbits were divided into two groups randomly,all the rabbits were contrasted to be models of angioplasty in iliac artery.After injury,the control group were feed with standard forage, the nesiritide group received standard forage plus subcutaneous nesiritide(0.1mg·kg~(-1)·d~(-1),20 days).The rabbits were killed 4 weeks after initial injury.The injured segments were processed for histological and morphological study by imumohistochemistry and TD 2000 analysis system of pathology image.Results There were significant increase in the indexes of AUIEM,AUEEM(P0.05),extremely significant difference between residual cavity areas,ratios of stenosis(P0.01). There was no significant difference between areas of media(P0.05).The expression of VEGF in endothelial cells and smooth muscle cells decreased in nesiritide group than control group.Conclusion It suggested that the nesiritide could benefit vascular remodeling after angioploasty,and inhibit the positive expression of VEGF.
