止哮汤对哮喘大鼠气道重塑及肺组织增殖细胞核抗原的影响

摘    要:目的观察止哮汤对哮喘大鼠气道重塑及肺组织增殖细胞核抗原(PCNA)的影响。方法选用健康Wistar大鼠60只,随机分为6组:正常对照组,哮喘模型组,地塞米松对照组,中药低剂量组、中剂量组、高剂量组。利用卵蛋白加氢氧化铝复制哮喘大鼠模型,各组大鼠均经末次激发24h后处死。取左肺在矢状面取材按常规方法作病理切片及HE染色。光镜下观察哮喘大鼠的气道改变;免疫组化法分别测定大鼠肺组织PCNA蛋白的表达。结果模型组大鼠气道壁及气道平滑肌面积明显增厚,气道壁面积、气道平滑肌面积以及支气管平滑肌细胞数经支气管内周长标准化后,亦明显高于对照组(P〈0.05)。正常对照组PCNA呈阴性表达,模型组PCNA表达呈强阳性,强于正常对照组(JP〈0.05);地塞米松组、中药各剂量组PCNA表达均弱于模型组;中药低剂量组强于地塞米松组(P〈0.05);中药中、高剂量组则与地塞米松组无明显差异(P〉0.05);中药中、高剂量组均弱于中药低剂量组(P〈0.05);中药高剂量组与中药中剂量组表达无明显差异(P〉0.05)。结论止哮汤可下调哮喘大鼠肺组织中PCNA的表达,减轻气道重塑,高、中剂量组疗效较好,与地塞米松效果相当。

Abstract:Objective: To observe Zhixiao Decoction on incidence of acute asthmatic rat Airway remodeling and lung tissue of PCNA. Method: 60 healthy Wistar rats were randomly divided into six groups: the normal control group, Zhixiao Decoction group, dexamethasone control group,low dose group, middle dose group, high-dose group six groups in total. Ovalbumin rat n~odel of asthma were made with aluminum hydroxide. The rats were sacrificed after the last challenge 24 h. Left lung in the sagittal plane drawn were taken, and histology and HE staining were made by conventional methods. Light microscope airway ch~nges in rats with asthma,acute asthma model was successfully verified;Immunohistochemical were measured in lung tissue PCNA protein. Results: Model rats air- way wall area and airway smooth muscle were thickening,airway wall area,airway smooth muscle area and the number of bronchial smooth muscle cells within the perimeter of the standardized,become higher. Normal group is negative,model group is positive and stronger than normal group;Dexamethasone group, Chinese PCNA expression in each dose group were weaker than the model group;Low dose group was stronger than in the dexamethasone group;Zhixiao Decoction,the high-dose group and dexamethasone group had no difference;Zhixiao Decoction,the high-dose group were lower than low dose group (all P〈 0.05) ;Chinese medicine and traditional Chinese medicine in the high dose group showed no difference in dose group. Conclusion: Zhixiao Decoction can reduced asthmatic lung tissue expression of PCNA and dexamethasone,high dose group good effect, and dexamethasone quite effective.
