pp242对膀胱癌T24细胞增殖及侵袭能力的影响

目的探讨pp242对膀胱癌T24细胞增殖和侵袭能力的影响。方法将T24细胞贴壁培养于10%胎牛血清、100 U/mL青霉素、100μg/mL链霉素的1640培养液中,37℃、5%CO2饱和湿度培养箱中培养。取对数生长期细胞,处理组加入浓度为50、100、200、500 nM pp242;对照组不加药物,每天更换1640培养液。采用CCK-8法检测各组细胞生长抑制率、transwell小室法检测细胞体外侵袭能力的变化。结果 pp242对T24细胞有显著的增殖抑制作用,transwell法检测pp242处理后的T24体外侵袭能力明显下降,均呈剂量依赖性。结论 pp242对膀胱癌T24细胞具有抑制增殖和侵袭能力的作用,有望成为一种新的膀胱癌治疗药物。

Effect of pp242 inhibiting proliferation and invasion of human bladder carcinoma cell line T24 in vitro

Objective To investigate the inhibiting effects of pp242 on proliferation and invasion of human bladder cancer cell T24.Methods Tumor cells were maintained in RPMI-1640 containing 10% fetal bovine serum,and penicillin/streptomycin.Cells were grown at 37 ℃ in a 5% CO2 incubator.T24 cells of logarithmic growth phase were treated with 50,100,200,500 nM concentrations of pp242 solution;cells in control group were without treatment.The tumor cell growth repression rate was measured by CCK-8 colorimetry.The invasion of T24 cells and the control cells were measured by transwell chamber assay.Results pp242 inhibited proliferation of T24 tumor cells in concentration-dependent manners.pp242 inhibited the invasiveness of T24 cells in transwell chamber assay.Conclusion pp242 inhibits growth and invasiveness of bladder carcinoma cells,and it might be a new drug to treat bladder cancer.
