四君子汤抑制移植性原发性肝癌小鼠肿瘤生长的实验研究

摘    要:目的 观察四君子汤对移植性原发性肝癌小鼠肿瘤生长的影响.方法 30只移植性原发性肝癌小鼠随机分为模型组、5 - FU组及四君子汤组(简称四君组),每组10只.模型组和四君组小鼠分别给予蒸馏水和四君子汤灌胃,每天1次,连续给药10 d;5 -FU组隔天给予5- FU腹腔注射.观察各组小鼠的体质量、瘤重、脾重、肿瘤细胞凋亡、细胞周期、表观扩散系数( ADC)及血清血管内皮生长因子(VEGF).结果 小鼠体质量和去瘤体质量比较,四君组显著高于其余两组(P<0.01).比较瘤重和肿瘤指数,四君组显著低于模型组(P<0.01).抑瘤率比较,5 - FU组(54.67%)>四君组(17.81%).脾脏重量及脾脏指数比较,四君组显著高于模型组和5 - FU组(P<0.01).细胞凋亡比较,四君组显著高于模型组(P<0.01).比较S期细胞,5 - FU组显著低于其余两组(P<0.01).G0 - G1期比较,四君组显著高于模型组(P<0.05),低于5 - FU组(P<0.05).G2 -M期各组比较差异无统计学意义(P>0.05).肿瘤ADC及血清VEGF比较,四君组低于模型组(P<0.01),高于5 - FU组(P<0.01).小鼠肿瘤ADC值与血清VEGF水平呈正相关(r=0.873,P<0.01),与细胞凋亡呈负相关(r=-0.284,P<0.05).结论 四君子汤对小鼠移植性原发性肝癌的作用机制在于促进肿瘤细胞的凋亡,影响细胞周期,抑制血清VEGF的表达;肿瘤实质的ADC值与实验室检查存在一定相关性,对于小鼠肝癌模型肿瘤生长情况的评定具有一定指导意义.

Experimental Study on Anti-tumor Effect of Sijunzi Decoction in Mice with Transplanted Primary Liver Cancer

Abstract:Objective To observe the effect of Sijunzi Decoction on tumor growth in mice with transplanted primary carcinoma of the liver. Methods 30 mice with transplanted primary carcinoma of the liver were randomly and equally divided into 3 groups,the blank control group consisted of untreated model mice,and 2 treated groups consisted of model mice treated with 5-FU(q.o.d,intraperitoneal injection,×5) and Sijunzi decoction(SJD)q.d.by gastrogavage for successive 10 days.Mice’s body mass,tumor size,spleen weight,tumor cell apoptosis and percentage of cells in different cell cycle stages,blood vascular endothelial growth factor(VEGF) level as well as the apparent diffusion coefficient(ADC) detected by magnetic resonance imaging(MRI) were observed. Results Among 3 groups,the heaviest body mass,de-tumor body mass,spleen index of mice were presented in the SJD treated group(P<0.01) while the tumor index of the SJD group is lower than the blank control group(P<0.01).Tumor inhibition rate and cell apoptosis rate in the 5-FU treated group was the highest and in SJD group the secondary.As for cells of different cell cycle stages,comparisons among the 4 groups showed that S phase cell in 5-FU group < SJD group < the blank control group(P<0.01);G0-G1 phase cell in the balnk control group < SJD group < 5-FU group(P<0.05 or P<0.01),G2-M phase cell showed no significant difference among them.Comparisons of ADC and VEGF level showed that 5-FU group < SJD group < the blank control group(P<0.01).The ADC value was correlation with the serum VEGF level(r=0.873,P<0.01)and tumor cell apoptosis(r=-0.284,P<0.05). Conclusion The acting mechanism of SJD on transplanted primary liver cancer may be accelerating tumor cell apoptosis and inhibiting serum VEGF expression.Change of MRI’s ADC value is keeping in step with changing of VEGF and apoptosis,thereby,it can objectively reflect the metabolism of tumor cells,being a new means for assessing the effect of Chinese medicine.
