电针对血管性痴呆小鼠海马神经元HSP70表达的影响

摘    要:目的:观察电针对血管性痴呆(VD)小鼠海马神经元凋亡及相关基因HSP70表达的影响。方法:以反复脑缺血再灌注法复制VD小鼠模型,电针肾俞、膈俞和百会穴,对照组灌胃喜得镇,分别于治疗1天和7天后,以FCM法检测小鼠海马细胞凋亡率、HSP70表达。结果:术后1天,模型小鼠海马细胞凋亡率明显增高,术后7天较术后1天还高。说明海马细胞由于反复脑缺血再灌注而出现凋亡。术后1天,模型小鼠海马细胞HSP70表达明显下降,7天时则显著升高。电针和喜得镇在两个时点均可上调小鼠海马细胞HSP70的表达,降低凋亡率,但以电针作用为优。结论:电针具有抑制细胞凋亡、保护神经元的作用,可能是电针改善VD小鼠智能的作用机理之一。

Abstract:Objective:To explore the effect of electropuncture on apoptosis and expressions of HSP70 in hippocampus neurons of mice with vascular dementia(VD).Methods:The VD model of mice was established by repeated cerebral ischemia-reperfusion,and were acupuncted the acupuncture points of shenshu,geshu,and baihui.The mice of control group were administered intragastrically with hydergine.With method of flow cytometry,to detect the apoptosis rate and expressions of HSP70 in hippocampus neurons of mice on the first and seventh day after treatment.Result: The apoptosis rate of hippocampus cells increased obviously on the first and seventh day after operation,which illustrated that hippocampus cells appeared apoptosis due to repeated cerebral ischemia-reperfusion.The expressions of HSP70 in hippocampus neurons decreased on the first day,but increased notablely on the seventh day.Electropuncture could up regulate the expressions of HSP70,lower the apoptosis rate of hippocampus cells of mice,which were more excellent than that of hydergine.Conclusion:The treatment with electropuncture could inhibit apoptosis,protect neurons,which possiblely is one of mechanism on improving intelligence of VD mice.
