Abstract:Objective To analyze the mechanism of the couplet medicine of Scrophulariae Radix-Ophiopogonis Radix in the treatment of anxiety disorders based on network pharmacology;To provide basis for clinical application of the couplet medicine of Scrophulariae Radix-Ophiopogonis Radix.Methods The chemical constituents of Scrophulariae Radix and Ophiopogonis Radix were collected by TCMSP and TCMIP.Active components were screened by TCMSP,SwissADME database and literature review.The target of active components was predicted based on SwissTargetPrediction database.The related target of anxiety disorder was screened through OMIM,GeneCards and DrugBank database.The protein-protein interaction(PPI)network of the common targets of active components and anxiety disorder was established through STRING database.Based on Cytoscape 3.8 software,MCODE cluster analysis was performed on PPI network to obtain the key targets.The DAVID 6.8 was used to perform gene ontology(GO)analysis and enrichment analysis of KEGG on the common targets of active components and anxiety disorder,and the core active components were screened through active components-target-pathway network.The binding strength of core active components and key targets was simulated by molecular docking.Results Totally 37 active components were screened out,including 471 potential targets,753 anxiety related targets and 42 common targets of active components-disease.Four types of clusters were obtained by MCODE cluster analysis of the PPI network,and 14 key targets were screened out.GO analysis and KEGG enrichment analysis showed that the treatment of anxiety disorder by the couplet medicine of Scrophulariae Radix-Ophiopogonis Radix mainly involved the molecular functions of GABA-A receptor activity,benzodiazepine receptor activity,serotonin binding,as well as the serotonergic synapse,the GABAergic synapse,the dopaminergic synapse and other 15 signaling pathways.Finally,12 core active components with strong binding strength to key targets and significant antioxidant and neural cell protection effects were screened out.Conclusion Couplet medicine of Scrophulariae Radix-Ophiopogonis Radix may regulate the activity of nerve cells through 5-HT,GABA receptor and DA and other targets and related chemical pathways,and treat anxiety disorders through mechanisms such as anti-oxidation,protection and promotion of nerve cell regeneration.
