血管生成素-1在小鼠肾脏发育中的表达及作用

摘    要:目的观察小鼠肾脏发育过程中血管生成素-1(Ang-1)的表达部位以及定量变化,探讨小鼠肾脏发育过程中Ang-1的作用及意义。方法分别取不同胚龄的胎鼠和不同日龄的仔鼠肾脏切片,进行HE和免疫组化染色及应用免疫印迹技术对Ang-1进行定量检测。结果免疫组化结果表明,Ang-1在胚龄14 d的胎鼠肾脏中开始出现表达,但表达微弱,部位在肾间质细胞。以后随着胚(日)龄的增加,表达逐渐增多且分布广泛,主要分布在肾小体、肾间质,肾小管以及肾血管中。免疫印迹结果表明,自胚龄14 d始,Ang-1蛋白含量逐渐增加,生后21 d达到最高水平,随后逐渐下降。结论在小鼠肾脏的发育的过程中,Ang-1表达部位及强度不同,表明其在小鼠肾脏发育以及成熟的过程中,调控和维持着正常肾小球滤过功能以及肾小管的回吸收功能。

Abstract:Purpose To observe expression parts and quantitative change of angiopoietin-1(Ang-1) during mouse kidney development in order to investigate the effect of Ang-1.Methods The kidneys of the foetus mice of different days and postnatal different days mice were taken.Kidneys of mice were used for observing the kidney development and the expression of Ang-1 by immunohistology and processed for Western blotting.Results The results of immunohistology showed that the expression of Ang-1 was faint at E14 and they were located in renal mesenchymal cells.Then the expression of Ang-1 tended to increase gradually following the kidney development and they were detected in glomeruli,blood vessels and renal tubules.The results of western blotting showed that the expression of Ang-1 protein increased gradually following the kidney development and peaked at 21 day,then decreased.Conclusion During mouse kidney development,Ang-1 was expressed in different region,and the intensities were evident.So it demonstrates that during the kidney development,Ang-1 is involved in nephrogenesis,growth and maturation.Simultaneously they have an important role in regulating and maintaining normal glomerular filtration.
