葛根异黄酮对去卵巢大鼠的抗骨质疏松作用及其机制

摘    要:目的研究葛根异黄酮对去卵巢大鼠抗骨质疏松的作用机制。方法选择3月龄健康雌性Wister大鼠60只,随机分为假手术组、模型组、尼尔雌醇组、葛根异黄酮低、中和高剂量组等6组,每组10只。假手术和模型组灌胃给予0.5%CMC-Na溶液;尼尔雌醇组灌胃给予尼尔雌醇0.5 mg/kg,每周2次(周一和周四);葛根异黄酮低、中和高剂量组分别灌胃给予葛根异黄酮25,50,和100 mg/kg,时间均为70 d。采用ELISA法测定血清细胞因子IL-6、IGF-1水平;免疫组化法测定颅骨成骨细胞骨形态发生蛋白-2(BMP-2)的表达;双能X线骨密度仪测量股骨中点及远心端骨密度。结果葛根异黄酮能明显的增加股骨中点及远心端骨密度(P<0.05),升高血清IGF-1的水平(P<0.05),降低血清IL-6的水平(P<0.01),并且能较好的增加成骨细胞BMP-2表达水平(P<0.01)。结论葛根异黄酮能够改善骨质疏松,作用机制可能通过下调IL-6表达水平和提高成骨细胞BMP-2的表达实现的。

Abstract:Purpose To investigate the mechanism of Pueraria Isoflavones on osteoporosis in ovariectomized rats.Methods 60 female Wister rats(three months) were divided into six groups randomly:sham operation group(0.5% CMC-Na),model group(0.5% CMC-Na),nilestriol group(0.5 mg/kg),Pueraria Isoflavones of low group(25 mg/kg),medium group(50 mg/kg) and high group(100 mg/kg).After 70 days of intragastric administration,ELISA,immunohistochemical technique and Dual-energy X-ray were used to assess the levels of serum cytokines IL-6,IGF-1,the expression of BMP-2 and the density of bone mineral in the central part and end part of the femur respectively.Results The experiment suggested that the Pueraria Isoflavone could increase the bone mineral density of bone mineral in the central part and end part of the femur significantly(P<0.05),elevate the level of serum IGF-1(P<0.05) and reduce the level of serum IL-6(P<0.01).Moreover,Pueraria Isoflavones could also increase the expression of BMP-2(P<0.01).Conclusion Pueraria Isoflavone can improve the symptoms of osteoporosis and the mechanism may be expressed by down-regulating IL-6 levels and improving bone cell BMP-2 expression.
