巴曲亭肺部给药抗新生大鼠急性肺出血作用的研究

目的研究巴曲亭从肺部给药途径抗新生大鼠急性肺出血中的治疗作用．方法出生7—10dSD大鼠随机分为5组（n=24）：（1）正常对照组；（2）巴曲亭气管滴入组（巴曲亭组，2．5Ku／kg）；（3）内毒素（lipopolysaccharide．LPS，5mg／kg）组，腹腔注射；（4）LPS（ip，5mg／kg）＋巴曲亭（2．5Ku／kg）气管滴入组（肺部给药组）；（5）LPS（ip，5mg／kg）＋巴曲亭（2．5Ku／kg）静脉注射组（静脉给药组）．用LPS（5mg／kg）腹腔注射复制新生SD大鼠急性肺出血模型．观测各组新生大鼠在给予巴曲亭后1h、2h和4h出血时间、凝血时间、凝血酶原时间和纤维蛋白原含量．结果与LPS组相比，巴曲亭肺部给药较静脉给药在同一时间点更能有效缩短急性肺出血新生大鼠的出血时间、凝血时间、凝血酶原时间，提高纤维蛋白原含量，减轻急性肺出血的程度．结论巴曲亭肺部给药在抗新生大鼠急性肺出血治疗作用上优于静脉给药．

Therapeutic Effect of Pulmonary Delivery of Hemocoagulaseon Acute Lung Hemorrhage Induced by Lipopolysaccharide inNeonatal Rats

Objective To investigate therapeutic effect of pulmonary delivery of hemocoagulase on acute lung hemorrhage induced by lipopolysaccharide in neonatal rats. Methods Healthy neonatal Sprague-Dawley rats were ramdomly divided into 5 groups： control, Hemorrhage, Lipopolysaccharidee delivery of hemocoagulas group, LPS ＋ venous delivery of hemocoagulas group, The group, LPS＋ pulmonary changes of bleeding （BT） , coagulation time （CT） , prothrombin time （PT） and fibrinogen were measured. All changes time were observed at 1 h, 2 h and 4 h after delivery of hemocoagulase. Results Compared with Lipopolysaccharide group, the lung hemorrahage was evidently ameliorated after pulmonary delivery of hemoeoagulase. Bleeding time, coagulation time and prothrombin time was shortened. Goaclusion Pulmonary delivery of hemocoagulase can shorten bleeding time and coagulation time of rats with acute lung hemorrhage.
