石榴皮总黄酮的微波提取工艺及含量测定研究

摘    要:目的:研究石榴皮中总黄酮的微波提取最佳工艺条件并测定其含量。方法:采用微波提取法提取石榴皮中总黄酮,通过单因素试验,考察了微波功率、乙醇体积分数、料液比、微波提取时间4个因素对总黄酮提取的影响,通过正交试验进一步优化提取工艺,运用硝酸铝显色法测定总黄酮含量。结果:微波提取法提取石榴皮中总黄酮的最佳工艺为乙醇体积分数60%,料液比为1∶25,微波提取时间为120 s;微波功率对总黄酮提取无显著影响,其余3个因素对总黄酮提取的影响依次为乙醇体积分数>料液比>微波提取时间;总黄酮平均含量为6.70%。结论:采用微波提取法提取石榴皮中的总黄酮,具有快速、高效、节能、不破坏有效成分等特点,具有良好的应用前景。

Abstract:Objective:To study the optimum process conditions of microwave extraction of total flavonoids in gra-nati pericarpium and determine the amount of total flavonoids in granati pericarpium . Methods: The microwave extrac-tion technique was used to extract the total flavonoids in granati pericarpium . The influence factors to the extraction were studied through single-factor tests , such as microwave power , ethanol concentration , material-to-liquid ratio and microwave extraction times . Through orthogonal tests the optimum conditions of microwave extraction were confirmed . The amount of total flavonoids was determined by aluminum nitrate assay . Results:The optimum conditions of mi-crowave extraction of total flavonoids in granati pericarpium were obtained including ethanol concentration of 60%, material-to-liquid ratio of 1∶25 and extraction time of 120 s . Microwave power had no significant influence on the total flavonoids extraction . The influence of the other three factors on the total flavonoids extraction was ethanol concentra-tion〉material-to-liquid ratio〉microwave extraction time . The content of total flavonoids in granati pericarpium was 6 . 70%. Conclusion:The microwave extraction of total flavonoids in granati pericarpiumis proved the advantages of high efficiency , energy saving and without destroying the effective components , and a good application prospect was showed .
