食管鳞状细胞癌组织中微卫星不稳定的研究

目的观察食管鳞状细胞癌组织中的微卫星不稳定（MSI）表达及临床意义。方法采用PCR技术和非变性聚丙烯酰胺凝胶电泳技术，检测45例食管鳞状细胞癌及相应癌旁正常组织2个位点的MSI状态。结果D3S1067位点MSI检出频率较高，为24．4％（11／45）；D18S58位点MSI阳性率为13．3％（6／45）。有3例既在D3S1067又在D18S58中检出MSI。有31对组织在D3S1067和D18S58两个位点均未检出MSI。MSI在食管鳞状细胞癌中表达率为31．1％（14／45），与肿瘤的病理病理分级、PTNM分期、有无区域淋巴结转移和侵润深度无关（p〉0．05）。结论食管鳞状细胞癌发生过程中存在MSI途径，MSI可能作为临床筛查食管鳞状细胞癌高危人群的分子手段，具有潜在的应用前景。

To investigate the expression andthe clinical signification of microsatellite instability in esophageal sqnamons cell carcinoma

Objective To investigate the expression and the clinical signification of microsatellite instability（MSI） in esophageal squamous cell 45pairs of esophageal squamous cell carcinoma and normal tissues were analyzed for the MSI expression （ two microsatellite lociwere detected） by PCR-based methods and denaturalized polyarylamide gel electrophoresis technique. Results MSI was the most frequently obs erved at D3S1067 with an incidence of 24.4% （11/45）; MSI at D18S58 was detected in 13.3% （6/45） of the tumors .31 pairs of tissues had no MSI at either D3S1067 or D18S58 .14 cases with MSI were observed in 45 esophageal squamous cell carcinoma. MSI at 3p and 18q has no sig nificant relationship with the tumor stage ,the cell differentiation ,the invasive depth of the tumor tissues and the regional lymph node metastasis （p 〉 0.05） .Conclusion In esophageal squamous cell carcinoma is associated with MSI, MSI appears to be a promising molecular index for the cli nical screening in the population at risk of esophageal squamous cell carcinoma.
