胸苷激酶基因治疗人脑胶质瘤SHG44的研究

摘    要:
构建了含有pgk启动子驱动的HSV-tk基因的反转录病毒载体pLNTK,并成功地转移至人脑胶质瘤SHG44细胞。体内实验证实,ACV可抑制SHGLNTK的肿瘤形成,并可抑制SHGLNTK肿瘤的生长。原位基因转移治疗SHG44肿瘤效果明显。提示其可作为脑肿瘤治疗的一种新方法。

Abstract:
RetroviralvectorpLNTKcaringHSV-tkgenedrivenbypgkpromoterwereconstructedandsucessfulytransferedintohumangliomacelSHG44.TheinvivostudyconfirmedthatACVcouldsuppresthetumorformationoftheSHGLNTKcelsandthegrowthofSHGLNTKtumors.ThisdemonstratedthatSHGLNTKtumorcelsweretreatedinsituwithinjectionretrovirussupernatantwithcontainedHSV-tkandimplantationofPA317celswhichproducedretroviralvectorpLNTK.There-sultsshowedthatsuchgenetransfertreatmentweremoreefectiveandindicatedthatsuchtreatmentmaybeusedasaninnovativemethodforbraintumortherapy.
