青藤碱对巨噬细胞吞噬能力的影响及机制探讨

摘    要:目的:观察青藤碱(sinomenine,SIN)对大鼠腹腔巨噬细胞吞噬能力的影响,并对其机制进行探讨.方法:大鼠随机分为对照组和青藤碱(20、60、180 mg/kg4组,酵母多糖A腹腔注射诱导急性腹膜炎,提取腹腔巨噬细胞,贴壁法纯化.鸡红细胞吞噬法测定巨噬细胞吞噬指数和吞噬率;分别采用荧光定量realtime RT-PCR和免疫组织化学方法检测TNF-α、IL-6 mRNA和蛋白表达.结果:20mg/kg青藤碱能明显提高巨噬细胞吞噬吞噬指数和吞噬率,明显降低巨噬细胞TNF-α、IL_6的表达:60、180mg/kg组能显著提高巨噬细胞吞噬指数和吞噬率,显著降低巨噬细胞TNF-α、IL-6的表达.结论:青藤碱能显著提高巨噬细胞吞噬能力,减少炎症因子表达.
