大鼠体内9-硝基喜树碱代谢物的HPLC-MSn分析

摘    要: 目的研究9-硝基喜树碱在大鼠体内的代谢物。方法静脉注射9-硝基喜树碱(6 mg·kg-1)12 h后,收集大鼠的胆汁、尿液和粪便。运用高效液相色谱和质谱联用技术检测9-硝基喜树碱的代谢产物。结果从给药后大鼠的胆汁、尿液和粪便中,发现并初步鉴定了6个主要代谢产物的化学结构。结论大鼠给予9-硝基喜树碱后,在胆汁中产生9-羟基喜树碱的葡萄糖醛酸结合物、9-乙酰胺基喜树碱、9-氨基喜树碱、9-羟基喜树碱及两个未知代谢产物;在尿液中的主要代谢产物为9-羟基喜树碱的葡萄糖醛酸结合物、9-乙酰胺基喜树碱、9-氨基喜树碱、9-羟基喜树碱;在粪便中的主要代谢产物为9-羟基喜树碱、9-乙酰胺基喜树碱及9-氨基喜树碱。

Abstract:OBJECTIVE To investigate the metabolites of 9-nitrocamptothecin in rats.METHODS After intravenous administration of 9-nitrocamptothecin(6 mg·kg-1)in rats,bile,urine and fecal excretion were collected over a 12 h period and the 9-nitrocamptathecin concentrations were determined by a HPLC-MSn method.RESULTS The major metabolites were found in rat bile,urine and fecal excretion and their chemical structures were identified primarily.CONCLUSION According to the rule of metabolism,the results indicate there are 6 metabolites in bile,4 metabolites in urine and 3 metabolites in fecal excretion.
