摘    要:目的:研究鹿角方的入血移行成分及规律,初步阐明其药效物质基础。方法:通过LC-MS/MS法分析大鼠含药血清,明确鹿角方的原型入血成分;及不同给药剂量下各移行成分相对峰面积与剂量的关系,解析移行规律。结果:通过与标准品保留时间、离子碎片信息的比对,鉴定鹿角方中13种成分(马钱苷、柚皮苷、橙皮苷、朝霍定A、朝霍定B、朝霍定C、淫羊藿苷、补骨脂素、异补骨脂素、宝霍苷Ⅰ、红景天苷、莫诺苷、特女贞苷)均能以原型入血;随给药剂量上升,柚皮苷、橙皮苷、补骨脂素、异补骨脂素、特女贞苷的相对峰面积与剂量呈线性关系,而马钱苷、莫诺苷、红景天苷、宝霍苷Ⅰ呈非线性关系,其中马钱苷、莫诺苷、补骨脂素、异补骨脂素、宝霍苷Ⅰ、红景天苷易入血。结论:马钱苷、莫诺苷、补骨脂素、异补骨脂素、宝霍苷Ⅰ、红景天苷可能是鹿角方体内直接作用的主要物质,实验结果为鹿角方药效物质基础的研究提供借鉴。

Abstract:OBJECTIVE To investigate migrating components absorbed into blood of Lujiao Formula (LJF) and their migrating regularity to preliminarily clarify the bioactive substances of LJF.METHODS The serum of rats and the relationship between the relative peak area(RPA) of each component and dosage were analyzed by LC-MS/MS to elucidate prototype components absorbed into blood of LJF as well as their migrating regularity.RESULTS By the comparison of the retention time and ion fission fragments with standards, thirteen characteristic components of LJF (loganin, naringin, hesperidin, epimedin A, epimedin B, epimedin C, icariin, psoralen, isopsoralen, baohuoside Ⅰ, salidroside, morroniside and specnuezhenide) were confirmed to be absorbed into blood in prototype form and their RPAs increased with increased dosage. The RPAs of naringin, hesperidin, psoralen, isopsoralen, and specnuezhenide at different dosage groups showed linear relationship with dosage, while loganin, morroniside, salidroside and baohuoside Ⅰ revealed non-linear relationship with dosage. Moreover, loganin, morroniside, psoralen, isopsoralen, baohuoside Ⅰ and salidroside could be easily absorbed into blood.CONCLUSION It is inferred that loganin, morroniside, psoralen, isopsoralen, baohuoside Ⅰ and salidroside might be the main substances of LJF which may play direct roles in vivo, which provide a basis for the study of the bioactive substances of LJF.
