铜绿假单胞菌慢性肺部感染大鼠的左氧氟沙星药动学研究

摘    要:
目的:建立铜绿假单胞菌(PA)慢性肺部感染大鼠模型,采用高效液相色谱法(HPLC)法测定不同剂量的左氧氟沙星(LVF)在其体内的血药浓度,并进行相关药动学(PK)研究。方法:建立PA肺部慢性感染大鼠模型,分别单剂量灌胃口服LVF 45.0 mg·kg-1和67.5 mg·kg-1,测定给药后的血药浓度和肺组织药物浓度,用DAS 2.0软件计算PK参数。结果:采用HPLC法测定PA慢性肺部感染大鼠生物样品中的LVF,该法回收率、准确度、精密度、线性范围及检测限均符合生物样品分析的要求。结论:LVF在PA慢性肺部感染大鼠体内的PK过程符合一级吸收二室模型。

Abstract:
OBJECTIVE To establish a rat model of chronic pulmonary infection with Pseudomonas aeruginosa (PA) and determine the plasma concentration of different doses of levofloxacin (LVF) by high performance liquid chromatography (HPLC).METHODS A rat model of PA pulmonary chronic infection was established.The rats were given a single dose oral administration of LVF of 45.0 or 67.5 mg·kg-1. Plasma and lung concentrations were determined at selected time points. The pharmacokinetic parameters were calculated by DAS 2.0 software.RESULTS HPLC method was used to determine LVF concentrations in the plasma or lung tissue. Recovery, accuracy, precision, linear range and detection limit of this method were all met the standard of biological sample analysis.CONCLUSION Pharmacokinetic of LVF in PA chronic pulmonary infectious model of rats was closely in line with the two-compartment model with first order absorption.
