摘    要:目的探讨85例乳腺浸润性导管癌中肿瘤干细胞标志物乙醛脱氢酶1(ALDH1)的表达情况及与临床病理特征的关系。方法采用免疫组化法检测乳腺组织芯片85例乳腺浸润性导管癌组织中ALDH1的表达并分析其临床意义。结果 ALDH1在雌激素受体(ER)阴性的乳腺癌中表达率(34.6%)显著高于ER阳性者(12.1%;P=0.024)。不同亚型乳腺癌中存在ALDH1表达的显著差异,其中三阴性乳腺癌中ALDH1的表达较高(50.0%,P=0.034)。ALDH1的表达在不同大小肿块的肿瘤中也存在差异,其中肿块小于2 cm的乳腺癌中ALDH1的表达率最高(50.0%;P=0.040)。而ALDH1的表达与患者年龄、肿瘤分级、淋巴结转移、疾病分期及PR、HER-2状态无关(P>0.05)。结论 ER阴性乳腺癌尤其是三阴性乳腺癌中ALDH1的表达率较高,提示ER阴性乳腺癌中存在较高比例的干细胞,进而有助于更好地理解其预后差的特点。

Expression of aldehyde dehydrogenase1(ALDH1) is associated with ER negative breast cancer

Abstract:Objective To investigate the expression of stem cell marker aldehyde dehydrogenase1(ALDH1) and its correlation with the clinical characteristics of breast invasive ductal cancer.MethodsWe performed immunohistochemistry to detect ALDH1 expression on breast cancer tissue microarray including 85 breast invasive ductal cancer tissues.Results 22 cases(25.9%) in the 85 breast invasive ductal cancer tissues were ALDH1 positive,though 17 of the 22 cases expressed ALDH1 at a low level.The expression frequency of ALDH1 in ER negative breast cancer was higher than that in ER positive breast cancer(34.6% vs 12.1%;P=0.024).ALDH1 expression differed among distinct subtypes of breast cancer,with a higher rate of positivity in triple-negative breast cancer(TNBC)(50.0%,P=0.034).Different expression of ALDH1 was also found in tumors with different sizes,among which tumors smaller than 2cm showed elevated expression of ALDH1(50.0%;P=0.040).Moreover,ALDH1 expression was not associated with age,tumor grade,lymph node status,stage,PR and HER-2 status(P>0.05).ConclusionsHigher expression of ALDH1 was mainly found in ER negative breast cancer,expecially in TNBC,suggesting a higher proportion of cancer stem cells in this kind of breast cancer.These data may be clinically useful for a better understanding of the poor prognosis of the patients with ER negative breast cancer.
