大肠癌中聚(腺苷二磷酸核糖)水解酶对聚(腺苷二磷酸核糖)聚合酶的调节作用及意义

摘    要:
目的 初步探讨人肠癌中聚(腺苷二磷酸核糖)水解酶[Poly(ADP-ribose)glycohydrolase,PARG]与聚(腺苷二磷酸核糖)聚合酶[Poly(ADP-ribose)polymerase,PARP]的关系.方法 应用S-P免疫绀化检测人大肠癌组织中PARG、PARP的表达;以PARG抑制剂丹宁酸(Gallotannin,GLTN)为处理因索,采用流式细胞计数检测小鼠结肠腺癌CT26细胞中PARG、PARP表达变化.结果 PARG和PARP在人大肠癌组织中的表达呈正相关(γ=0.300 01,P<0.05);流式细胞计数显示,用GLTN处理后,小鼠结肠腺癌CT26细胞中PARG、PARP的阳性细胞率分别较对照组降低,差异具有统计学意义(P<0.05).结论 大肠癌中PARG和PARP的表达有相关性,PARG抑制剂可以抑制PARG及PARP表达,PARG叮能对PARP具有调节作用.
