摘    要:目的:研究盐酸左氧氟沙星脂质体的包封率测定方法和影响因素,初步考察其体外释放规律.方法:用硫酸铵梯度法制备了盐酸左氧氟沙星脂质体;用凝胶过滤色谱法测定脂质体的包封率,并研究各种因素对包封率的影响;用溶出度第三法考察了脂质体的释放规律.结果:制得脂质体的包封率约为80%,脂质体的体外释放规律符合一级动力学过程.结论:体系pH值和硫酸铵浓度对包封率影响最大,制得脂质体的体外释放具有缓释特点.

Abstract:AIM:To build a determination method of encapsulation efficiency(EE)for levofloxacin liposome,to investigate the factors affecting EE,and to study the release profile in vitro.METHOD:Levofloxacin liposome was prepared by ammonium sulfate gradient method.The EE was determined by gel permeation chromatography to investigate the effect of some factors on EE.The release behavior of the drug from liposome was studied by the third method for dissolution.RESULT:The EE of liposome reached 80%,and the release rule in vitro was subject to the first order model.CONCLUSION:The pH and concentration of ammonium sulfate were the most important two parameters to EE.The drug was sustained-released from the liposome.
