蛇床子素对人黑色素瘤A375细胞增殖和凋亡的影响

摘    要:目的:探讨蛇床子素对人黑色素瘤A375细胞增殖和凋亡的影响。方法:人黑色素瘤细胞株A375细胞体外培养,将对数生长期的A375细胞以不同浓度的蛇床子素处理,MTT法检测A375细胞增殖;流式细胞术检测A375细胞凋亡;RTPCR法检测A375细胞Bcl-2 mRNA和Bax mRNA表达水平;Western blotting检测A375细胞IκB-α、p-NF-κB、NF-κB、Bcl-2和Bax蛋白表达。结果:蛇床子素可降低A375细胞增殖率以及Bcl-2 mRNA、p-NF-κB和Bcl-2蛋白表达水平,呈浓度依赖性;同时增加A375细胞凋亡率以及Bax mRNA、IκB-α、Bax蛋白表达水平,呈浓度依赖性,但对NF-κB表达无明显影响。结论:蛇床子素能诱导A375细胞增殖抑制和凋亡,呈浓度依赖性,其机制可能与抑制NF-κB信号通路激活有关。

Abstract:Objective:To investigate the effect of Osthole in proliferation and apoptosis of human melanoma cell lines A375.Methods:The human melanoma cell lines A375 were cultured in vitro and after the A375 cell at logarithmic growth phase was treated with different concentration of Osthole.The proliferation of A375 cell was detected by methyl thiazolyl tetrazolium (MTT) assay;the flow cytometry was used to determine the apoptosis of A375.RT-PCR was used to detect the mRNA level of Bcl-2 and Bax in A375;Western blotting was used to detect the expression of I-κB,p-NF-κB,NF-κB,Bcl-2 and Bax in human melanoma cell line A375.Results:Osthole can decrease the proliferation rate,the mRNA level of Bcl-2 and the expression of p-NF-κB and Bcl-2 protein level in human melanoma cell line A375 in a concentration dependent manner.Moreover,Osthole can increase the apoptosis of A375 cells and the mRNA level of Bax and the expression of IκB-α and Bax protein level and had no influence in the expression of NF-κB.Conclusion:Osthole could induce the proliferation inhibition and apoptosis of A375 cell and the effect is associated with suppressing NF-κB signal transduction pathway activation.
