半夏泻心汤联合川乌对胃溃疡大鼠血清胃泌素水平的影响

摘    要:目的:观察半夏泻心汤加减半夏反药川乌对胃溃疡大鼠血清胃泌素水平的影响。方法:SD大鼠50只,随机抽出10只作为正常组,其余大鼠禁食不禁水24 h后,以冷水浸法诱导应激性大鼠胃溃疡模型,将造模成功大鼠随机分为模型组、半夏泻心汤组及半夏泻心汤加川乌组。于次日开始给药,正常组及模型组给予生理盐水10 m L·kg~(-1)灌胃,半夏泻心汤组给予半夏泻心汤水煎液10 m L·kg~(-1)灌胃,半夏泻心汤加川乌组给予半夏泻心汤10 m L·kg~(-1)及川乌水煎液10m L·kg~(-1)灌胃,连续给药7 d,记录各组大鼠摄食量及体质量变化,末次给药后,大鼠禁食12 h,眼眶采血,分离血清,以酶联免疫法检测大鼠血清胃泌素水平,分离大鼠心、肝、脾、肺、肾、胃、胸腺等脏器并称质量,以精密p H试纸检测大鼠胃液p H值,并以游标卡尺测量溃疡面积和溃疡抑制率。结果:半夏泻心汤及半夏泻心汤加川乌组大鼠体质量比给药前有所升高,其中半夏泻心汤加川乌组大鼠体质量高于半夏泻心汤组;与模型组比较,半夏泻心汤组及半夏泻心汤加川乌组大鼠摄食量增加,胃液p H值升高(P0.05),溃疡面积及溃疡抑制率降低(P0.01);此外,半夏泻心汤加川乌组大鼠血清胃泌素水平显著升高,肾脏系数显著降低,与正常组比较,有显著性差异(P0.05)。结论:半夏泻心汤加川乌可升高胃溃疡大鼠血清胃泌素水平,且对胃溃疡大鼠肾脏有一定损伤。

Abstract:Objective:To investigate the effect of Banxia Xiexin Decoction and Radix Aconiti on Serum Gastrin in Rats with Gastric Ulcer.Method:A total of 50 SD rats were employed in the study.10 rats were randomly selected as the normal control group,while other 40 rats were fasted yet had free access to water for 24 hours.Then the gastric ulcer model rats were induced by water-immersion stress method.The gastric ulcer model rats were randomly divided into model group,Banxia Xiexin Decoction group and the Banxia Xiexin Decoction plus Radix aconite group.On the second day after modeling,the normal control group and the model control group were administrated with normal saline at dose of 10 mL · kg-1,and the Banxia Xiexin Decoction group were administrated with Banxia Xiexin Decoctionat at dose of 10 mL · kg-1.The Banxia Xiexin Decoction plus Radix aconitig roup were administration with Banxia Xiexin Decoction and Radix aconiti at dose of 10 mL · kg-1,respectively.Seven days later,all rats' body weight and food intake were observed.After the last administration,all rats were fasted for 12 hours.The the blood samples were collected from eyes and the serum was separated by centrifugation.The expression of gastrin in serum was tested with enzymelinked immunosorbent assay,and the organ coefficients of heart,liver,spleen,lung,kidney,stomach and thymus were investigated.The gastric fluid pH was determined with accurate pH test paper,and the ulcer area and ulcer inhibition rate were measured with a digital caliper.Results:There were rising tendencies of body weight in Banxia Xiexin Decoction treated group and Banxia Xiexin Decoction plus Radix aconiti group,in which the effect of Banxia Xiexin Decoction was better.Compared with the model group,the food intake and PH value increased (P < 0.05) and the ulcer area and ulcer inhibition rate decreased (P < 0.01) in Banxia Xiexin Decoction group and Banxia Xiexin Decoction plus Radix aconite group.Moreover,compared with normal control group,the expression of gastrin in serum was increased (P < 0.05) and the kidney coefficient decreased (P < 0.05) significantly in Banxia Xiexin Decoction plus Radix aconite group.Conclusion:Banxia Xiexin Decoction plus Radix aconiti could best increase the expression of gastrin in serum and exhibited side effect on the kidney of the gastric ulcer model rats.
