建立大鼠失神经支配骨骼肌萎缩模型适宜方法的研究

目的：探讨建立大鼠失神经支配骨骼肌萎缩动物模型的适宜方法。方法：选取雄性Wistar大鼠48只随机分为两组,无菌条件下,分别切断左后肢坐骨神经和左后肢胫神经。术后观察动物的生活状态,并于术后1、2、4、8周分别测定手术侧与健侧腓肠肌的肌湿重、肌纤维横截面。结果：术后动物全部存活,但切断坐骨神经组有3只动物手术侧肢体远端出现皮肤溃疡。切断坐骨神经或胫神经2周后,大鼠肌湿重、肌纤维横截面积的测定值,手术侧与健侧相比较即出现显著性差异,术后4周内肌萎缩进程较快,随后趋缓。结论：建立失神经支配骨骼肌萎缩的动物模型,两种方法比较,切断坐骨神经对动物损害较大,建议采用切断胫神经这种方法较为适宜。

Study on Proper Methods of Amyotrophy Models of Denervated Skeletal Muscle in Rat

Objective: To compare methods of setting up animal model of denervated skeletal muscle atrophy in rats.Methods: Forty-eight male Wistar rats were used and randomly divided into 2 groups.Rats in group A were cut off the left sciatic nerve under aseptic conditions.Rats in group B were cut off the left tibial nerve under aseptic conditions.Health level of animals after operation were observed.Muscle wet weight and cross-sectional area of gastrocnemius were detected in 1,2,4,8 weeks after operation.Results:All rats were alive after operation.Among the rats which had been cutting off the sciatic nerve,there were 3 rats appeared to skin canker beyond leg.By comparing the values of the muscle wet weight and cross-sectional area in the damaged and healthy parts of each individual,the difference in both group A and B were statistically significant(P<0.05) since 2 weeks after operation.The amyotrophy process was faster in 4 weeks after operation.Conclusions:By compare the harmness to animal,the method of cutting off the tibial nerve is more suitable than that of cutting off the sciatic nerve in order to set up the model of denerved skeletal muscle in rat.
