血管性痴呆大鼠海马神经细胞内游离钙含量的长时程变化

摘    要:目的观察血管性痴呆(VD)大鼠海马神经元细胞内静息态游离钙离子含量([Ca^2+]i)的长时间变化规律。方法将60只大鼠随机分为假手术组、模型7d组、模型15d组、模型1m组、模型2m组和模型4m组,每组10只。在激光共聚焦显微镜下显微荧光测钙法(钙荧光探针Fulo-3/AM)检测海马组织的静息态钙荧光强度。结果各模型组钙荧光强度均明显高于假手术组(P〈0.01);模型各组间比较,海马神经细胞静态钙荧光强度有显著性差异(P〈0.01)。而且随着造模后时间的延长,钙荧光强度逐渐增加。结论VD大鼠海马神经元内游离钙离子含量明显升高,随脑缺血再灌后时间的延长升高程度更加明显。

Abstract:Objective To observe the long term changes of intracellular free calcium concentration(i) of hippocampal neurons in resting state in vascular dementia rats.Methods VD rat models were established.Sixty rats were randomly divided into the sham-operated group(SO),7-day VD model group(7dM),15-day VD model group(15dM),1-month VD model group(1mM),2-month VD model group(2mM) and 4-month VD model group(4mM).The [Ca^2+]i of hippocampal neurons of each group rats,indicated by fluorescent value,were detected by fluorescent microscopy(used the Fulo-3/AM as calcium fluorescent indicator) under confocal laser scanning microscope.Results The Fluorescent value of resting [Ca^2+]i of hippocampal neurons was markedly higher in the model groups than in SO group(P〈0.01).The fluorescent values of resting [Ca^2+]i of hippocampal neurons among the model groups were significantly different(P〈0.01) and were gradually increased along with the time prolongation after the cerebral ischemia/reperfusion.Conclusion The [Ca^2+]i of hippocampal neurons in resting state was gradually elevated after cerebral ischemia/reperfusion in vascular dementia rats.
