白花丹醌一单壁碳纳米管偶联物的制备及其对人肝癌Bel-7402细胞的抑制作用

摘    要:
目的:制备白花丹醌单壁碳纳米管(plumbagin-singlewalledcarbonnanotubesconjugate,PLB-SWNT)偶联物,考察其在水溶液中的分散性和体外对人肝癌Bel一7402细胞的抑制活性。方法:以聚乙二醇(polyethyleneglycol,PEG)作为偶联剂,通过酰胺化和酯化反应将PLB与SWNT偶联,制备PLB-SWNT偶联物,并利用傅里叶转换红外光谱仪和透射电子显微镜(TEM)考察和分析所制备得到的偶联物的组成、结构和形貌,以紫外可见分光光度法(uV—Vis)测定PLB-SWNT偶联物PLB含量,并以MTT比色法、HE染色法和TEM等方法对该偶联物体外抑制Bel-7402活性进行初步研究。结果:①制备得到在水溶液中分散性较好的PLB-SWNT偶联物;②PLB-SWNT偶联物能够抑制人肝癌Bel-7402细胞的增殖。结论:①以PEG和PLB对SWNT进行共价修饰,可以制备分散性良好的PLB—SWNT偶联物;②PLB-SWNT偶联物在体外对人肝癌Bel-7402细胞的增殖抑制效果显著。

Abstract:
Objective:To synthesize plumbagin-single walled carbon nanotube conjugate (PLB-SWNT), and to investigate its dispersibility in water and inhibitiory effect on human hepatocarcinoma Bei-7402 cells in vitro. Methods: Conjugating plumbagin and single walled carbon nanotubes by amidation and esterification reaction was used by using polyethylene glycol (PEG) as coupling agent. Components, structure and mor- phology of the prepared conjugate were investigated and analyzed by using Fourier transform infrared spec- troscopy (FT-IR) and transmission electron microscope (TEM). The inhibitory effect of conjugates on Bel- 7402 was tested in vitro by MTT assay, HE staining and TEM observation. Results: The as-prepared PLB- SWNT conjugate exhibited high water dispersibility. PLB-SWNT conjugate could inhibit the proliferation of hepatocarcinoma Bel-7402 cells. Conclusion: The highly dispersive PLB-SWNT conjugate in water could be obtained after functioning SWNTs by using PEG and PLB. The inhibitory effect of PLB-PEG-SWNTs on the proliferation of tumor cells in vitro was remarkable.
